摘  要:目的探究羧基麦芽糖铁(ferric carboxymaltose,FCM)及蔗糖铁(iron saccharate,IS)对HL-7702正常肝细胞的毒性及氧化应激作用。方法选用浓度为2、1、0.5、0.25、0.125、0.062 mg·mL^(-1)的静脉铁剂与HL-7702细胞共培养24 h,MTT法和乳酸脱氢酶(LDH)释放法测定2种静脉铁剂对HL-7702细胞毒性作用,比色法检测细胞中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、还原性谷胱甘肽(GSH)以及脂质过氧化产物丙二醛(MDA)的活力和含量。结果2种静脉铁剂分别对HL-7702细胞培养24 h后,高浓度组细胞存活率与对照组比较显著降低。2种静脉铁剂与HL-2207细胞孵育24 h后,高浓度IS组的LDH释放率显著高于对照组及FCM组;高浓度FCM组的LDH释放率显著高于对照组。SOD、CAT活力下降,MDA含量增加,均与静脉铁剂浓度呈正相关。结论结果表明FCM及IS对HL-7702细胞的毒性作用均与其浓度呈正相关,且对HL-7702细胞的脂质过氧化及氧化应激毒性与其发挥细胞毒性有相关性。不同静脉铁剂对HL-7702细胞发挥致死性作用的剂量不同,IS的细胞毒性大于FCM。Objective In this study, the cytotoxicity and oxidative stress of Ferric Carboxymaltose(FCM) and Iron Saccharate(IS) on HL-7702 cells was observed.Methods HL-7702 cells were co-cultured with intravenous iron at concentrations of 2,1,0.5,0.25,0.125 and 0.062 mg·ml^(-1) for 24 hours.The cytotoxic effects of two intravenous chalybeatus on HL-7702 cells were determined by MTT and LDH release methods.The activity and content of superoxide dismutase(SOD),catalase(CAT),glutathione(GSH) and malondialdehyde(MDA),a lipid peroxidation product, were detected by colorimetry.Results After 24 hours of co-culture of HL-7702 cells with two intravenous chalybeatus, the cell survival rate was significantly lower than that of control group.After 24 hours incubation of HL-2207 cells with two intravenous chalybeatus, the LDH release rate of IS group was significantly higher than that of control group and FCM group, and the LDH release rate of FCM group was significantly higher than that of control group.Increased dose was positively correlated with intravenous iron concentration.Conclusion The results showed that the toxicity of FCM and IS to HL-7702 cells was positively correlated with their concentration, and the oxidative stress of HL-7702 cells was correlated with their cytotoxicity.Different intravenous iron agents had different doses of lethal effects on HL-7702 cells, and the cytotoxicity of IS was higher than that of FCM.
