摘  要:目的:研究白藜芦醇(Res)调控调节性T细胞/辅助性T细胞17(Treg/Th17)抑制溃疡性结肠炎(UC)肠道黏膜炎症机制。方法:50只小鼠随机分为模型对照组、Res低剂量组、Res高剂量组、阳性对照组和阴性对照组,每组各10只。模型对照组、Res组以及阳性对照组小鼠使用5%葡聚糖硫酸钠(DSS)建造急性UC模型,阴性对照组为正常小鼠,Res组小鼠在建立了UC模型后用不同剂量白藜芦醇处理,阳性对照组用柳氮磺吡啶栓处理。收集小鼠的结肠黏膜组织,提取淋巴细胞测定Treg和Th17在T淋巴细胞的占比以及Treg/Th17比值并检测外周血炎症因子的表达。结果:与阴性对照组比较,模型对照组小鼠Treg细胞占比、Treg/Th17下调;Th17细胞占比,IL-17、IL-23、IL-1β、IL-6和TNF-α上调(P<0.05);与模型对照组比较,Res低剂量组、Res高剂量组以及阳性对照组Treg细胞占比、Treg/Th17上调;Th17细胞占比,IL-17、IL-23、IL-1β、IL-6和TNF-α下调(P<0.05)。且高剂量组对上述指标影响程度更大。结论:Res能够调控Treg/Th17免疫失衡,抑制UC的肠道炎症反应。Objective:To study the mechanism of resveratrol(Res)regulating Treg/Th17 to inhibit intestinal mucosal inflammation in ulcerative colitis(UC).Methods:50 mice were randomly divided into Model control group,Res low-dose group,Res high-dose group,Positive control group and Negative control group,with 10 mice in each group.The Model control group,Res group and Positive control group were treated with 5%dextran sodium sulfate(DSS)to establish the acute UC model,while the Negative control group was normal mice,the Positive control group was treated with sulfasalazine suppository.The proportion of Treg and Th17 in T lymphocytes,the ratio of Treg/Th17 and the expression of inflammatory factors in peripheral blood were detected.Results:Compared with the Negative control group,the proportion of Treg cells,Treg/Th17 was down-regulated;the proportion of Th17 cells,and IL-17,IL-23,IL-1β,IL-6 and TNF-αwere up-regulated in the Model control group(P<0.05),Compared with the Model control group,the proportion of Treg cells,Treg/Th17,and Th17 cells in the Res low-dose group,Res high-dose group and Positive control group were up-regulated;and IL-17,IL-23,IL-1β,IL-6 and TNF-αwere down-regulated(P<0.05).And the high-dose group has a greater impact on the above indicators.Conclusion:Res can regulate the immune imbalance of Treg/Th17 and inhibit the intestinal inflammatory response of UC.
