摘 要:目的研究黄芩苷对糖尿病大鼠心肌保护机制。方法2019年7月采用SD大鼠尾静脉注射45 mg/kg链脲佐菌素建立实验性糖尿病模型,将40只大鼠按照随机区组设计法分为对照组(NC组,腹腔注射生理盐水)、糖尿病模型组(DM组,腹腔注射生理盐水)、黄芩苷+糖尿病组[Baicalin+DM组,腹腔注射黄芩苷药液15μmol/L/(kg·d)]和黄芩苷+对照组[Baicalin+NC组,腹腔注射黄芩苷药液15μmol/L/(kg·d)],每组10只。用药8周后检测各组大鼠血糖、心脏重量指数等,masson染色观察各组大鼠心肌组织变化,计算心肌胶原容积分数(CVF)。采用Western-blotting测定白细胞介素-1β(IL-1β)、肿瘤坏死因子α(TNF-α)、细胞Toll样受体4(TLR4)、核因子-κBp65(p65)、Ⅰ型胶原(collagenⅠ)蛋白水平。结果与DM组比较,Baicalin+DM组大鼠血糖、心脏重量指数均明显降低,差异均有统计学意义(P<0.05)。DM组大鼠心肌细胞排列紊乱,肌纤维间胶原纤维增生明显;Baicalin+DM组大鼠心肌细胞排列较DM组规则,胶原纤维增生明显减少,CVF明显降低,差异均有统计学意义(P<0.05)。DM组大鼠IL-1β、TNF-α、TLR4、p65、collagenⅠ蛋白水平均较NC组明显升高;与DM组比较,Baicalin+DM组大鼠IL-1β、TNF-α、TLR4、p65、collagenⅠ蛋白水平明显降低,差异均有统计学意义(P<0.05)。结论黄芩苷通过抑制TLR/核因子κB信号通路减轻心肌炎性反应,抑制胶原纤维增生对糖尿病大鼠心肌具有保护作用。Objective To study the myocardial protective mechanism of baicalin in diabetic rats.Methods In July 2019,SD rats were injected with 45mg/kg streptozotocin via tail vein to establish experimental diabetes models,40 diabetes rats were randomly divided into control group(NC group,intraperitoneal injection of normal saline),diabetes mellitus group(DM group,intraperitoneal injection of normal saline),baicalin+diabetes mellitus group[Baicalin+DM,intraperitoneal injection of Baicalin solution 15μmol/L/(kg·d)]and Baicalin+control group[by intraperitoneal injection of Baicalin+NC group,Baicalin solution 15μmol/L/(kg·d)],with 10 rats in each group.Eight weeks later,blood glucose and cardiac mass index were detected.Masson staining observed myocardial tissue changes in each group,and collagen volume fraction(CVF)was calculated.Western-blotting detected IL-1β,TNF-α,TLR4,p65 and collagenⅠ.Results Compared with DM groups,blood glucose and cardiac mass index were decreased in Baicalin+DM groups(P<0.05).The myocardial cell was disorganized and the collagen fibers between the muscle fibers proliferated obviously by masson staining method in DM groups.Myocardial cell was organized and collagen fibers hyperplasia decreased obviously of Baicalin+DM groups,CVF was decreased,with statistical significance(P<0.05),Western-blotting showed that IL-1β,TNF-α,TLR4,p65 and collagenⅠin DM groups were significantly higher than those in NC groups.Compared with DM groups,IL-1β,TNF-α,TLR4,p65 and collagenⅠwere decreased in Baicalin+DM groups,with statistical significance(P<0.05).Conclusion Baicalin has protective effect on myocardium of diabetic rats by inhibiting TLR/NF-κB signaling pathway,reducing myocardial inflammatory response and inhibiting collagen fiber proliferation.
