摘 要:目的:探讨采用非离心的免疫磁珠吸附去除红细胞(ErythroClear),快速检测冷冻复苏后微量脐带血的造血功能的效果。方法:选取2014年11月—2020年12月在广州脐血库冷冻复苏后的脐带血样本(附属血辫),分为洗涤离心去除红细胞的常规法组(76例)与免疫磁珠吸附去除红细胞的新方法组(73例)。观察两种方法在样本制备质量和速度方面的差异,评价脐血冷冻前(大袋取样)和复苏后(附属血辫取样)的总有核细胞数、细胞活率、CD34阳性细胞数及祖细胞集落数等质量指标及变化情况(回收率)。结果:新方法组的细胞活率的回收率低于常规法组[(79.11±9.06)%vs.(84.63±4.17)%],差异有统计学意义(P<0.05),而两组的总有核细胞回收率、CD34阳性细胞回收率,粒-巨噬细胞集落回收率及祖细胞集落回收率的比较,差异均无统计学意义(P>0.05)。结论:免疫磁珠法可用于快速检测冷冻复苏后脐带血的造血功能,特别适用于冷冻袋附属血辫的微量样本。Objective To investigate the method of depleting red blood cells(RBCs)by centrifuge-free immunomagnetic beads(ErythroClear)from small volume samples in rapidly detecting the hematopoietic function of cryopreserved cord blood units after thaw.Methods From November 2014 to December 2020,the attached segments of cryopreserved cord blood units after thaw in Guangzhou cord blood bank were selected as the research objects and divided into wash-centrifuged RBCs depletion group(named as regular method group,76 cases)and immunomagnetic-bound RBCs depletion group(named as new method group,73 cases).The differences on quality and efficiency of sample processing between the two groups were observed,and the quality parameters of the cord blood samples before(sampling from bulk)and after cryopreservation(sampling from attached segment)were assessed,including the total nucleated cells(TNCs),cell viability,CD34 positive cells,colony forming units-granulocyte/macrophage(CFU-GMs)and colony forming units(CFUs),as well as the change of the above parameters(recovery rate).Results The recovery rate of cell viability in the new method group was lower than that in regular method group[(79.11±9.06)%vs.(84.63±4.17)%].The difference was statistically significant(P<0.05).There was no significant difference in the recovery rate of TNCs,CD34 positive cells,CFU-GMs and CFUs between the two groups(P>0.05).Conclusion Immunomagnetic-based method can be used for rapid detecting the hematopoietic function of cryopreserved cord blood units after thaw,especially for the small volume segments attached to the freezing bags.
