载多柔比星骨水泥复合体体外药物释放的实验研究

摘  要:目的探讨多柔比星/骨水泥复合体及多柔比星/阿仑膦酸钠/骨水泥复合体中多柔比星的释放规律及阿仑膦酸钠对多柔比星释放的影响。方法分别制备多柔比星/骨水泥复合体及多柔比星/阿仑膦酸钠/骨水泥复合体,各组随机抽取6个试样,置于烧杯中,加入60 mL磷酸盐缓冲液,37℃孵育。分别于不同时间点(1 h、2 h、4 h、6 h、8 h、12 h、24 h、2 d、3 d、5 d、7 d、10 d)取1 mL试样液,每次取液后补充1 mL乙腈-甲醇溶液(3∶1),测定多柔比星药物浓度,绘制时间与累积释放曲线。结果两组复合体体外释放在0~24 h有明显突释效应,24 h后有缓释效应,第5 d达到药物浓度峰值。两组在不同时间点的多柔比星释放浓度比较,差异无统计学意义(P>0.05)。结论多柔比星在聚甲基丙烯酸甲酯(PMMA)骨水泥中能够稳定、持续地释放,并存在突释效应,阿仑膦酸钠不影响多柔比星的释放。Objective To investigate the release of doxorubicin from doxorubicin-loaded bone cement and doxorubicin-alendronate-loaded bone cement and the effects of alendronate on the release of doxorubicin. Methods Doxorubicin-loaded bone cement and doxorubicin-alendronate-loaded bone cement were prepared respectively. Six samples from each group were randomly selected and incubated in a beaker with 60 mL phosphate buffer at 37 ℃. Each time 1 mL solution was taken out and 1 mL acetonitrile-methanol mixture(3∶1) was added, and the sample solution was taken at different time points(1 h, 2 h, 4 h, 6 h, 8 h, 12 h, 24 h, 2 d, 3 d, 5 d, 7 d, 10 d) to determine the drug concentration of doxorubicin released. The time and cumulative release curves were drawn. Results Both groups showed an obvious burst release in vitro at 0~24 h, and then a sustained release after 24 h, reaching a concentration peak at the 5th day. There was no significant difference in doxorubicin-released concentration between the two groups at different time points(P>0.05). Conclusion Doxorubicin can be released stably and continuously in polymethyl methacrylic(PMMA) bone cement, and it has a burst release initially. Alendronate had no effects on the release of doxorubicin.
