摘 要:为阐明细胞壁降解酶活性与种子萌发之间的相互关系,以莴苣种子为材料,研究了内切-β-甘露聚糖酶、α-半乳糖苷酶和β-甘露糖苷酶在莴苣种子萌发不同阶段的活性变化。结果表明,莴苣种子吸胀曲线呈“S”型,种子于18 h时开始萌发,随后萌发率迅速上升,48 h时达95.11%;干种子、吸胀种子、狭义萌发种子和完全萌发种子中细胞壁降解酶活性存在显著差异,随着萌发进程的推进,内切-β-甘露聚糖酶、α-半乳糖苷酶活性不断增加,其中前者增加最为显著,而β-甘露糖苷酶活性变化则呈递减趋势。莴苣种子在萌发过程中可通过增加细胞壁降解酶活性来降解细胞壁甘露聚糖,从而弱化胚乳,促进种子萌发。In order to clarify the relationship between cell wall degrading enzyme activity and seed germination,the activity changes of three major degrading enzymes of cell wall,i.e.endo-β-mannanase,α-galactosidase andβ-mannosidase,were studied in different stages of lettuce seed germination.The results showed that the imbibition curve of lettuce seed was“s”type.Lettuce seed began to germinate at 18 h,then the germination rate rose rapidly,and the germination rate was 95.11%at 48 h.There were significant differences in degrading enzyme of cell wall activities among dry seeds,bloated seeds,narrow sense germination seed and complete germination seed.With the advance of germination,the activities of endo-β-mannanase andα-galactosidase both increased continuously,the former increased most significantly,while the activity ofβ-mannosidase decreased.This indicated that the mannan of cell wall could be degraded by increasing the activity of cell wall degrading enzyme during the process of lettuce seed germination,so as to weaken endosperm and promote seed germination.
