摘  要:本研究旨在探讨苦木注射液(KM)对阿奇霉素(AZM)在肺炎模型大鼠体内组织分布的影响。将肺炎模型大鼠随机分为对照组和实验组,分别经尾静脉注射AZM和/或KM,剂量均为75 mg/mL,每日1次,连续给药1周。LC-MS/MS法测定末次给药24 h后大鼠血浆和组织中AZM浓度,比较对照组与实验组AZM组织分布差异。结果显示,建立的LC-MS/MS方法经考察符合生物样品分析要求,适用于AZM浓度测定。与对照组相比,实验组血浆、脾、肺和肾组织中AZM浓度没有显著性差异,心和肝中的AZM浓度显著降低(P<0.05)。研究表明KM对AZM在大鼠心和肝中的分布具有显著影响,这可能是通过KM促进AZM在体内代谢而降低其组织蓄积,有利于避免产生肝损伤。This study aims to investigate the effect of Kumu injection(KM)on the tissues distribution of azithromycin(AZM)in rats with pneumonia.Pneumonia model rats are randomly divided into a control group and an experimental group.AZM and/or KM are injected via tail vein at a dose of 75 mg/mL,once a day for consecutive week.LC-MS/MS method is used to determine the concentration of AZM in the plasma and tissues of rats 24 h after the last administration,and to compare the distribution of AZM in the control and experimental group.The results show that the established LC-MS/MS method meets the requirements of biological sample analysis and is suitable for the determination of AZM concentration.Compared with the control group,AZM in the experimental group has no significant difference in plasma,spleen,lung and kidney tissues,but obviously decreases in heart and liver tissues(P<0.05).It is shown that KM has a significant impact on the distribution of AZM in heart and liver of rats.It may be that KM promotes the metabolism of AZM in body,thereby reducing the accumulation of AZM in the tissues,which is beneficial to avoid the risk of liver injury.
