摘 要:目的基于网络药理学探讨痛经宁方治疗子宫内膜异位症的分子机制。方法将通过中药系统药理学数据库与分析平台(TCMSP)得到痛经宁方的有效活性成分,与根据OMIM数据库和GeneCards数据库搜索得到的子宫内膜异位症疾病靶点交集,取得交集靶点。使用Cytoscape 3.7软件构建“中药-活性成分-靶点”网络。基于STRING数据库,构建蛋白互作(PPI)网络。利用DAVID数据库,进行痛经宁方潜在作用靶标的GO功能富集分析和KEGG通路富集分析。结果从痛经宁方中筛选出活性成分166个,其中61个有效活性成分可通过60个靶点作用于子宫内膜异位症。通过富集得到1177个GO生物功能和118条KEGG通路。结论基于网络药理学初步探究痛经宁方中槲皮素、山柰酚、花生四烯酸、异鼠李素、豆甾醇、β-谷甾醇、鞣花酸等有效成分作用于IL-6、VEGFA、EGFR、MAPK8、CASP3、ESR1等关键靶点,通过调控机体内类固醇激素水平、炎症反应、氧化应激、核受体活性、转录因子活性、细胞凋亡等过程治疗子宫内膜异位症,为进一步研究其具体作用机制提供了理论依据。Objective To explore molecular mechanism of Tongjingning recipe in the treatment of endometriosis based on network pharmacology.Methods The effective active ingredients of Tongjingning prescription were obtained through TCMSP,and the disease targets of endometriosis were obtained according to the OMIM database and Genecards search to obtain the intersection targets.The network of“traditional Chinese medicine-active ingredients-target”was constructed with the software of Cytoscape 3.7.Protein interaction(PPI)network was constructed based on string database.Using David database,the enrichment analysis of go function and KEGG pathway of potential targets of tongjingningfang were carried out.Results 166 active components were screened from Tongjingning prescription,61 of which could act on endometriosis through 60 targets.1177 GO biological functions and 118 KEGG pathways were obtained by enrichment.Conclusion Based on the network pharmacology,the effective components of Tongjingning prescription,such as quercetin,kaempferol,arachidonic acid,isorhamnetin,stigmasterol,β-sitosterol and ellagic acid,act on the key targets such as IL-6,VEGFA,EGFR,mapk8,CASP3,ESR1,etc.,by regulating the level of steroids,inflammatory response,oxidative stress,nuclear receptor activity,transcription factor activity,and cells Apoptosis and other processes in the treatment of endometriosis provide a theoretical basis for further study of its specific mechanism.
