摘  要:目的探讨以喷砂表面改性技术制备出的钛合金种植体对成骨细胞黏附、增殖及分化的影响。方法选取Ti-6Al-4V钛合金作为制备原材料,以喷砂表面改性技术制备出喷砂处理钛合金种植体实验标准片。实验分为两组,一组采用喷砂表面改性处理记为喷砂处理组,一组采用抛光处理记为Ti-6Al-4V对照组。利用扫描电镜检测两组钛合金种植体样品表面。利用原代细胞培养方法培养兔成骨细胞,将成骨细胞接种至两组钛合金种植体样品表面。扫描电镜观察接种3 d和7 d的细胞形态。MTT法检测细胞接种后1,4,7 d的细胞毒性以及增殖活性。结晶紫染色法检测细胞接种12 h和24 h的黏附情况。检测细胞接种后1,4,7 d的碱性磷酸酶(ALP)活性。结果扫描电镜检测发现喷砂处理组钛合金种植体样品表面较Ti-6Al-4V对照组粗糙。扫描电镜观察细胞接种后3 d和7 d的细胞形态以及结晶紫染色检测细胞接种后12 h和24 h的结果显示喷砂处理组表面的细胞OD值高于Ti-6Al-4V对照组(P<0.05),提示喷砂处理组的细胞黏附效果优于Ti-6Al-4V对照组。MTT法检测结果显示喷砂处理组在细胞接种后1,4,7 d的细胞相对增殖率分别为(91.98±1.21)%,(95.09±1.62)%以及(97.07±1.72)%;Ti-6Al-4V对照组在细胞接种后1,4,7 d的细胞相对增殖率分别为(88.17±1.13)%,(91.03±1.33)%及(93.56±1.67)%;两组之间细胞相对增殖率差异有统计学意义(P<0.05),提示两组钛合金种植体实验片的细胞毒性分级均为0级,并且喷砂处理组的成骨细胞增殖率高于Ti-6Al-4V对照组。ALP检测结果显示喷砂处理组细胞接种后1,4,7 d的ALP活性均高于Ti-6Al-4V对照组(P<0.05),提示喷砂处理组的成骨分化活性高于Ti-6Al-4V对照组。结论喷砂表面改性技术制备出的钛合金种植体对成骨细胞的黏附、增殖以及分化均具有促进作用。Objective To investigate the effect of titanium alloy implants prepared by sandblasting surface modification technology on the adhesion,proliferation and differentiation of osteoblasts.Methods Ti-6Al-4V titanium alloy was consecutively treated by sandblasting surface modification technology.The experiment was divided into two groups.Ti-6Al-4V titanium alloy was treated with sandblasting surface modification in sandblasting treatment group.Ti-6Al-4V titanium alloy was given polishing treatment in Ti-6Al-4V control group.Scanning electron microscopy was employed to observe the surface morphology of the prepared specimens.Rabbit osteoblasts cultured by the primary culture method were seeded on each specimen.Scanning electron microscopy was employed to observe the surface morphology of cells 3 d and 7 d after the inoculation.MTT assay was used to quantitatively determine the cytotoxicity and the proliferation of osteoblasts on each specimen co-cultured for 1,4,7 d.Osteoblasts were seeded on each specimen and co-cultured for 12 h and 24 h.The crystal violet staining was used to detect the adhesion of cells on the surface of materials.Alkaline phosphatase(ALP)of osteoblasts on the prepared specimen was detected at 1,4,7 d after inoculation.Results Scanning electron microscopy results indicated that the surface of the prepared specimens in sandblasting treatment group was rougher than that in Ti-6Al-4V control group.Scanning electron microscopy results at 3 d and 7 d and the crystal violet staining at 12 h and 24 h showed that the OD value in sandblasting treatment group was higher than that in Ti-6Al-4V control group(P<0.05),indicating that the adhesion of osteoblasts in sandblasting treatment group was better than that in Ti-6Al-4V control group.MTT assay results showed that the relative proliferation rates of osteoblasts in sandblasting treatment group were(91.98±1.21)%,(95.09±1.62)%and(97.07±1.72)%at 1,4,7 d.The relative proliferation rates of osteoblasts in Ti-6Al-4V control group were(88.17±1.13)%,(91.03±1.33
