墓头回药材中总黄酮和总皂苷测定方法研究

摘　　要：目的建立紫外-可见分光光度法测定墓头回中总皂苷与总黄酮。方法以齐墩果酸为对照品采用香草醛乙酸-高氯酸比色法进行显色反应测定总皂苷;以芦丁为对照品、亚硝酸钠-硝酸铝-氢氧化钠为显色体系,进行显色反应测定总黄酮,确定最优显色条件并进行相应的方法学考察。结果墓头回总皂苷的最佳显色条件为:反应温度80℃、显色时间20 min、1%香草醛乙酸-高氯酸溶液加入量0.5 ml;总黄酮最佳显色条件是硝酸铝溶液加1.5 ml、静置时间25 min、显色温度20℃。墓头回总皂苷与总黄酮在该显色条件下,方法学良好,符合药典规定。结论建立的紫外-可见分光光度法测定墓头回总皂苷与总黄酮方法简便,结果可靠,可以为墓头回中总皂苷与总黄酮的进一步研究提供参考。Objective To establish an ultraviolet-visible spectrophotometric method for determining total saponins and total flavonoids in Patrinia heterophylla Bunge subsp.heterophylla.Methods Taking oleanolic acid as the reference substance,the total saponins were determined by the color reaction of vanillin-perchloric acid colorimetry.Taking rutin as the reference substance,the total flavonoids were determined by sodium nitrite-aluminum nitrate-sodium hydroxide colorimetry.The optimal color reaction conditions were screened and the corresponding methodological investigation was carried out.Results The best color reaction conditions of total saponins from Patrinia heterophylla Bunge subsp.heterophylla were as follows:reaction temperature of 80℃,color development time of 20 min,and 1%vanillin acetic acid-perchloric acid solution of 0.5 ml.The best color reaction conditions of total flavonoids were 1.5 ml aluminum nitrate solution,standing time of 25 min and color development temperature of 20℃.The methodology of total saponins and total flavonoids of Patrinia heterophylla under the chromogenic condition was approved and met the requirements of the Pharmacopoeia.Conclusion The established ultraviolet-visible spectrophotometric method for the determination of total saponins and total flavonoids in Patrinia heterophylla Bunge subsp.heterophylla is simple and reliable,which could provide reference for the further study on total saponins and total flavonoids.
