摘  要:目的对高危型人乳头瘤病毒51型(HPV-51)L1蛋白的B细胞表位进行预测和分析。方法从Swissprot蛋白质数据库中检索HPV51型L1蛋白序列,采用生物信息学软件分析其理化性质、二级结构、柔韧性、亲水性、表面可及性、抗原性等参数,运用ExPASy网站的TMPRED和PROTSCALE软件分析预测其跨膜区域;采用氨基酸抗原性指数方法计算肽段的平均抗原指数(AI),将得到的肽段序列用Protein BLAST进行在线同源性匹配分析。结果 HPV51型L1蛋白由504个氨基酸残基组成,分子质量单位为56.31487 ku,等电点8.38。预测人乳头瘤病毒51型L1蛋白的B细胞表位可能位于其N端的5-10、51-57、75-96、124-132、138-152、174-185、214-217、230-234、269-286、316-320、336-341、355-360、428-443、475-486、489-497肽段。采用氨基酸抗原性指数方法计算AI,筛选出10条HPV 51型L1蛋白的B细胞优势表位。同源性分析显示75-96、138-152、174-185、428-443区段为可能的HPV51型L1蛋白优势B细胞表位。结论生物信息学方法预测HPV51型L1蛋白含有多个B细胞表位优势区域,可为其表位疫苗的研究提供理论基础。Objective To predict and analyze the B-cell epitopes of the L1 protein of high-risk human papillomavirus type 51(HPV-51). Methods The L1 protein sequence of HPV-51 was obtained from the Swissprot protein database. Bioinformatic software was used to analyze its parameters, such as its physical and chemical properties, secondary structure, flexibility, hydrophilicity, surface accessibility, and antigenicity. The software TMPRED and PROTSCALE from the ExPASy website were used to analyze and predict its transmembrane area. The average antigen index(AI) of peptide fragments was calculated based on the antigenicity of amino acids. Finally, the homology of the peptide fragment sequences was analyzed with the online software Protein BLAST. Results The L1 protein of HPV-51 consists of 504 amino acid residues, with a relative molecular weight of 56.31487 ku and an isoelectric point of 8.38. The transmembrane prediction indicated that the L1 protein likely to be a soluble protein with no transmembrane regions. B-cell epitopes of the L1 protein of HPV-51 were predicted to be located at the peptide fragments 5-10, 51-57, 75-96, 124-132, 138-152, 174-185, 214-217, 230-234, 269-286, 316-320, 336-341, 355-360, 428-443, 475-486, and 489-497 of the N-terminus. Ten dominant B-cell epitopes of the L1 protein of HPV-51 were selected according to the AI, which was calculated using Wu’s method. Further analysis of the sequence homology of the above peptide fragments indicated that the peptide fragments 75-96, 138-152, 174-185, and 428-443 were potential dominant B-cell epitopes. Conclusion Bioinformatic prediction indicated that the L1 protein of HPV51 contains multiple dominant B-cell epitopes, and this finding could provide a theoretical basis for further research on epitope vaccines against HPV51.
