摘  要:目的:初步探究盐酸千金藤碱(CH)对人牙髓干细胞(DPSC)增殖和成骨分化能力的影响。方法:体外分离培养鉴定人DPSC,MTT法检测0.0、2.5、5.0、10.0和20.0μmol/L CH处理24、48和72 h对DPSC增殖能力的影响,Western blot检测DPSC中骨钙素(OCN)和核心结合因子(RUNX2)的表达。结果:DPSC细胞为典型的间充质干细胞形态,表达间充质干细胞表面标记物CD44且有成骨分化能力。2.5和5.0μmol/L CH可促进DPSC的增殖且使DPSC中OCN及RUNX2蛋白的表达水平升高(P<0.05)。结论:CH可促进人DPSC的增殖和成骨分化的能力。Aim:To preliminarily investigate the effects of cepharanthine hydrochloride(CH)on the proliferation and osteogenic differentiation of human dental pulp stem cell(DPSC).Methods:Human DPSC were isolated,cultured and identified from permanent teeth in vitro.MTT assay was used to evaluate the proliferation of DPSC treated by CH at different concentrations(0.0,2.5,5.0,10.0 and 20.0μmol/L)and time durations(24,48 and 72 h).The expressions of osteocalcin(OCN)and runt-related transcription factor-2(RUNX2)of DPSC were assessed by Western blot.Results:DPSC were typical mesenchymal stem cells which expressed CD44,and had the potential of osteogenic differentiation.CH at 2.5 and 5.0μmol/L could both enhance the proliferation of DPSC and the expression levels of OCN and RUNX2(P<0.05).Conclusion:CH can enhance the proliferation and osteogenic differentiation of human DPSC.
