摘 要:人胚胎干细胞(hESC)可以分化成包括间充质干细胞(MSC)在内的各种体细胞。尽管目前hESC体外衍生MSC(hESC-MSC)细胞的方案多种多样,但是它们的衍生效率相差较大,特别是对hESC-MSC细胞产物的免疫调节特性仍存在争议。文章针对h ESC-MSC细胞的免疫调节特性进行了探索,建立了简易、高效的hESC-MSC细胞制备方案。结果表明:hESC-MSC细胞比人脐带间充质干细胞(UC-MSC)具有更强烈的对CD4^(+)T淋巴细胞增殖的抑制和促进Treg分化的免疫调节功能,提示h ESC-MSC细胞可成为更有效的细胞制剂运用于治疗免疫相关的病症。Human embryonic stem cells(hESC) can differentiate into almost all types of cells in the body, including mesenchymal stem cells(MSC). Despite of intensive research on the generation of hESC-derived MSC(hESC-MSC), with various efficiency, it has been controversial for the immunomodulatory function in hESC-MSC cells. We established a simple and high-efficient protocol for the hESC differentiation towards MSC, and investigated the immunomodulatory properties of such hESC-MSC. The results showed that this hESC-MSC had significant effects on CD4^(+)T lymphocyte proliferation inhibition and Treg differentiation promotion;its immunomodulatory function was stronger than that of umbilical cord derived mesenchymal stem cells(UC-MSC). This hESC-MSC had superior immunomodulatory effects on CD4^(+)T lymphocytes, implying that it might be well suitable for the treatment in immune-related diseases.
