摘 要:【目的】探索沙冬青种子总黄酮对小鼠免疫器官淋巴细胞活性的影响.【方法】将50只试验小鼠随机分为空白组、免疫抑制组、黄酮Ⅰ组、黄酮Ⅱ组及黄酮Ⅲ组,每组10只,灌胃给药.试验28 d后,分别测定脏器指数(胸腺、脾脏)并采集样品进一步采用石蜡包埋切片和染色技术、免疫组织化学分析技术等,探讨胸腺、脾脏中淋巴细胞活性的变化.【结果】小鼠免疫抑制模型复制成功,CTX显著诱导小鼠免疫器官淋巴细胞凋亡.同时,不同剂量沙冬青种子总黄酮灌胃试验组,能不同程度促使免疫抑制小鼠脾脏和胸腺淋巴细胞增殖,进而促使胸腺皮质和脾脏白髓面积增大,尤其总黄酮灌胃剂量150 mg/(kg•d)组小鼠表现最明显,胸腺皮质和脾脏白髓面积分别达到(83.11±1.25)μm^(2)、(47.00±1.55)μm^(2),显著高于免疫抑制组(P<0.01).免疫组化分析显示,不同剂量沙冬青种子总黄酮灌胃试验组,能不同程度促使免疫抑制小鼠脾脏和胸腺CD3阳性T淋巴细胞及CD20阳性B细胞增殖,尤其在总黄酮灌胃剂量150 mg/(kg•d)组增殖效果最突出,均显著高于免疫抑制组(P<0.01).【结论】沙冬青种子总黄酮可显著促进免疫抑制小鼠T、B淋巴细胞增殖,修复受损免疫器官,从而改善和增强机体免疫功能.【Objective】To investigate the effect of total flavonoids from the seeds of Ammopiptanthus mongolicus on the lymphocyte activity in immune organs of mice.【Method】A total of 50 Kunming mice were randomly divided into 5 groups(n=10,half male and half female):normal control group,immunosuppression group,test groupⅠ,test groupⅡand test groupⅢ,followed by intragastric administration of medicine to mice.After 28 days,thymus index and spleen index were measured,and the tissue samples were collected.Subsequently,the lymphocyte activity in immune organs of mice was determined by the ways of paraffin embedding microtomy and staining techniques and immunohistochemical analysis techniques.【Result】The immunosuppressed mice were replicated successfully with CTX(i.e.cyclophosphamide),which could distinctly induce the apoptosis of lymphocyte in spleen and thymus.Compared with the immunosuppression group,the treatment by intragastric administration of total flavonoid could to different extent contribute to the proliferation of lymphocytes in thymus and spleen of immunosuppressed mice,and enlarge the area of thymic cortex and white pulp of spleen.Especially,in test group[i.e.total-flavonoids administration:150 mg/(kg·d)],the area of both organ regions were(83.11±1.25)μm^(2) and(47.00±1.55)μm^(2),respectively,which were significantly larger than the immunosuppression group(P<0.01).By immunohistochemical analysis,it was indicated that the treatments with total flavonoids could significantly promote the proliferation of both CD3 positive T and CD20 positive B lymphocytes in the spleen and thymus,which was especially distinct in test groupⅡ,compared with the immunosuppression group.【Conclusion】It was suggested that the total flavonoids from the seeds of A.mongolicus can promote the proliferation of lymphocyte T and B in thymus and spleen of immunosuppressed mice,and also can enhance their immune function by repairing the damaged immune organs.
