摘  要:【目的】构建流动注射化学发光的2种Luminol体系检测重楼中总皂苷的含量.【方法】基于Luminol-H_(2)O_(2)和KMnO_(4)-Luminol-H+两种体系能产生化学发光,加入重楼皂苷后可抑制体系的发光强度.因此,构建流动注射化学发光检测重楼中总皂苷的分析方法,同时开展了方法验证和干扰试验,并对两种体系分别进行了流程及条件优化.在优化条件下,Luminol-H_(2)O_(2)和KMnO_(4)-Luminol-H+两种体系的相对发光强度分别与重楼皂苷Ⅶ浓度在一定范围内呈良好线性关系.【结果】该方法的检出限分别为0.2015μg/mL和0.1021μg/mL,平均回收率分别为99.96%和97.23%;检测样品中重楼总皂苷平均得率分别为21.138 mg/g和21.317 mg/g.【结论】所构建的两种化学发光体系,其线性检测范围宽、检出限低、精密度好,可用于重楼药材的品质分析和含量检测.【Objective】To construct two kinds of Luminol systems by CL-FIA for the determination of total saponins in Paris polyphylla var.chinensis.【Method】A flow injection chemiluminescence method for the detection of total saponins in P.polyphylla var.chinensis was established based on the chemiluminescence intensity of the Luminol-H_(2)O_(2)and KMnO_(4)-Luminol-H+which could be inhibited by adding the polyphyllins,and the method verification and interference tests were carried out,the flow paths and conditions of the two systems were optimized.There was a good linear relationship between the relative luminescence intensity of Luminol-H_(2)O_(2)and KMnO_(4)-Luminol-H+and the concentration of the polyphyllinsⅦin a certain range with the optimized conditions.【Result】The detection limits of the method were 0.2015μg/mL and 0.1021μg/mL,the average recoveries were 99.96%and 97.23%,the average yield of the total saponins in the samples was 21.138 mg/g and 21.317 mg/g.【Conclusion】The method with wide linear detection range,low detection limit and good precision can be used for the quality analysis and content detection of P.polyphylla var.chinensis.
