摘  要:目的研究去氧鬼臼毒素在大鼠和人血浆蛋白的结合率。方法建立去氧鬼臼毒素LC-MS/MS测定法,测定透析内、外液中2种成分质量浓度,并计算血浆蛋白结合率。结果去氧鬼臼毒素线性关系、精密度与准确度、提取回收率和稳定性均符合方法学要求。去氧鬼臼毒素在高、中、低浓度(500,1500、4500ng·mL^(-1))中,大鼠血浆蛋白结合率分别为(94.91±0.33)%、(94.52±0.50)%、(94.39±0.75)%,人血浆蛋白结合率分别为(93.88±0.46)%、(93.31±0.76)%、(93.13±0.60)%。结论LC-MS/MS法用于去氧鬼臼毒素生物样品测定具有简便、灵敏与选择性高的特点;去氧鬼臼毒素与人和大鼠血浆都具有高蛋白结合率,但二者无显著性差异。OBJECTIVE To determine the protein binding rate of deoxypodophyllotoxin in human and rat plasma.METHODS LC-MS/MS method was used to determine the binding rates in human and rat plasma of deoxypodophyllotoxin with dialysis method.RESULTS The linearity,precision and accuracy,extraction recovery and stability of deoxypodophyllotoxin all met the methodology requirements.Within the low,middle and high concentration(500,1500、4500 ng·mL^(-1))of deoxypodophyllotoxin,the protein binding rate of rat plasma was(94.91±0.33)%,(94.52±0.50)%and(94.39±0.75)%,respectively.While the protein binding rate of human plasma was(93.88±0.46)%,(93.31±0.76)%and(93.13±0.60)%,respectively.CONCLUSION Deoxypodophyllotoxin have high protein binding rate both in human and rat plasma.however,no significant difference between the two kinds of plasma.
