摘  要:目的探讨大鼠下丘脑重要代谢产物变化,为功能性消化不良(FD)临床治疗提供依据。方法24只SD雄性大鼠用随机数字表分为对照组、模型组、针刺组各8只。模型和针刺组制作FD动物模型,对照组自然喂养。针刺组取胃俞募穴电针刺激10天,对照和模型组仅束缚。干预后检测血清胃泌素,以氢质子磁共振波谱成像检测下丘脑肌醇、氨基丁酸、肌酸等绝对浓度。结果与对照组比较,模型组一般情况较差,体重日增长量、进食量、饮水量和胃排空率明显降低,胃泌素水平增高,下丘脑肌醇、丙氨酸降低;针刺组较模型组,一般情况好转,体重日增长量、进食与饮水量、胃排空率明显升高,胃泌素水平降低,下丘脑肌醇升高,差异有统计学意义(P<0.05)。结论胃俞募穴可明显调节FD大鼠胃排空率和胃泌素,改善疾病状态,可能与其靶向性调节下丘脑代谢产物肌醇和丙氨酸有关。Objective To investigate the changes of important metabolites in the hypothalamic(HYP)zone of functional dyspepsia(FD)model rat in order to provide scientific basis for FD clinical treatment.Methods Twenty-four male SD rats were randomly divided into control group,model group and acupuncture group,8 animals in each group.The FD model was created in the acupuncture and model groups.The control group was fed naturally.Eelectroacupuncture was performed at Shu-Mu points of the stomach for 10 days in the acupuncture group while the other two groups were only bound.After intervention,levels of serum gastrin(GAS)were measured.The absolute concentrations of hypothalamic metabolites such as myo-inositol(Myo),γ-aminobutyric acid(GABA)and creatine(Cr)in hypothalamus were also measured by using hydrogen proton magnetic resonance spectroscopy(1H-MRS).Results Compared to the control group,general condition,daily weight gain,food intake,water intake and gastric emptying rate were significantly decreased and the concentrations of GAS was increased while Myo and Ala were reduced in hypothalamus of model group.Compared with the model group,the general condition was improved and daily weight gain,food intake,water intake and gastric emptying rate were increased and levels of GAS were increased while Myo and alanine(Ala)in hypothalamus were decreased(P<0.05).Conclusion Electroacupuncture at Shu-Mu points of the stomach can regulate the gastric emptying rate and GAS level and improve the general conditions.This may be related to its targeted regulation of hypothalamic metabolites such as Myo and Ala concentrations.
