摘　　要：为探究Dactinosclerus actinosclerus RM菌株的分泌蛋白的氨基酸组成和信号肽序列的特点,基于该菌株的全基因测序结果,利用生物信息学知识和软件对其编码的基因进行了分析.结果表明:RM菌株有3869个基因,其中439个基因编码的蛋白质含有信号肽,这些蛋白质中含侧链基团较小的氨基酸残基(如丙氨酸、亮氨酸、甘氨酸、苏氨酸和缬氨酸)的数量多,占总数摩尔比49.72%;含侧链基团较大的氨基酸残基(如酪氨酸、甲硫氨酸、色氨酸和组氨酸)的数量少,占总数摩尔比为4.37%;RM菌株有321个胞外分泌型蛋白,84个分泌型膜蛋白.这些胞外分泌蛋白的信号肽中,富含疏水性氨基酸残基(摩尔比为63.65%),而带正电荷(摩尔比为9.15%)和负电荷(摩尔比为1.15%)的残基较少,信号肽的切割位点序列主要为X-A-X-A型.胞外分泌蛋白中含有10个(6种类型)的蛋白酶,这些蛋白酶的信号肽氨基酸残基组成基本相似,不以蛋白酶种类差异而具有差异.In order to investigate the composition of amino acids and the characteristics of signal peptide sequences in the secreted proteins of Deinococcus actinosclerus RM strain,the genes of RM strain were analyzed by bioinformatics knowledge and software based on the whole gene sequencing results.The results showed that the RM strains coded 3869 genes,and 439 genes encoded proteins with signal peptide.The number of amino acid residues containing small side chain groups(such as alanine,leucine,glycine,threonine and valine)in these proteins was large,accounting for 49.72%of the total,and the number of amino acid residues containing large side chain groups(such as tyrosine,methionine,tryptophan and histidine)was small,accounting for 4.37%of the molar ratio.Further,analysis showed that RM strain had 321 extracellular proteins and 84 secretory membrane proteins.The signal peptides of these extracellular proteins were rich in hydrophobic amino acid residues(molar ratio,63.65%),positively charged(molar ratio,9.15%)and negatively charged(molar ratio,1.15%)were few.The cleavage site sequence of signal peptide is mainly X-A-X-A type.There were 10 extracellular proteases(6 types):the amino acid residues of the signal peptide of these proteases were basically similar,and did not vary according to the type of protease.
