摘  要:目的:探讨电针对带状疱疹后神经痛(PHN)大鼠星形胶质细胞激活的抑制及一氧化氮(NO)/诱导型一氧化氮合酶(iNOS)转导通路的影响。方法:制备PHN大鼠模型,随机分为模型组、阳性药物组、电针组、假电针组,另设对照组,每组有大鼠25只。对照组、模型组腹腔注射等体质量生理盐水,阳性药物组灌胃卡马西平,电针组电针阳陵泉穴、环跳穴,假电针组针刺上述两穴。实验后1~5周检测各组大鼠痛觉行为学,免疫荧光组织化学染色观察计数脑星形胶质细胞及胶质纤维酸性蛋白(GFAP)表达,实验第5周后检测脑组织NO含量和iNOS活性,采用逆转录-聚合酶链式反应法和蛋白质印迹法测定大鼠脑组织iNOS mRNA及其蛋白表达。结果:阳性药物组和电针组实验后1~5周热痛阈、机械痛阈持续升高,模型组、假电针组热痛阈、机械痛阈持续降低,阳性药物组、电针组高于模型组(P<0.001)。实验后1~5周对照组大鼠脑星形胶质细胞未激活,其余4组均激活。与模型组比较,实验后3~5周阳性药物组与电针组大鼠星形胶质细胞计数较少(P<0.001);与阳性药物组比较,电针组脑星形胶质细胞计数较多(P<0.001)。与模型组比较,电针组、阳性药物组大鼠脑组织GFAP荧光染色密度较小,NO含量及iNOS活性减小,iNOS mRNA及iNOS蛋白表达均较低(P<0.001)。结论:电针可抑制PHN大鼠脑星形胶质细胞的激活,可能通过抑制NO表达与iNOS释放,减轻大鼠疼痛感。Objective:To investigate the inhibition of electroacupuncture on the activation of astrocytes and nitric oxide(NO)/inducible nitric oxide synthase(iNOS)transduction pathway in rats with postherpetic neuralgia(PHN).Methods:PHN models were prepared and randomly divided into model group,positive drug group,electroacupuncture group,sham electroacupuncture group,and control group was set up.There were 25 rats in each group.The control group and the model group were intraperitoneally injected with the same volume of normal saline,the positive drug group was given carbamazepine intragastrically,the electroacupuncture group was electroacupuncture Yanglingquan(GB34)and Huantiao(GB30)points,and the sham electroacupuncture group was acupuncture the above two points.One to five weeks after the experiment,the pain behavior of rats in each group was detected.The expression of astrocytes and glial fibrillary acidic protein(GFAP)was observed and counted by immunofluorescence histochemical staining.The content of NO and activity of iNOS in brain tissue were detected.The expression of iNOS mRNA and protein in the brain tissue were determined by reverse transcription-polymerase chain reaction and Western blotting.Results:The thermal pain threshold and mechanical pain threshold of the positive drug group and the electroacupuncture group continued to increase 1 to 5 weeks after the experiment,the thermal pain threshold and mechanical pain threshold of model group and sham electroacupuncture group continued to decrease,the positive drug group and electroacupuncture group is higher than the model group(P<0.001).Brain astrocytes were not activated in the control group,but were activated in the other 4 groups.Compared with the model group,the astrocyte count of the positive drug group and the electroacupuncture group was less 3 to 5 weeks after the experiment(P<0.001);Compared with the positive drug group,the electroacupuncture group had higher cerebral astrocyte count(P<0.001).Compared with the model group,the GFAP fluorescence staining
