五加生化胶囊对早孕大鼠药物流产出血干预作用的代谢组学研究

摘　　要：目的:运用代谢组学方法探讨五加生化胶囊对早孕大鼠药物流产出血治疗作用及其机制。方法:以米非司酮配伍米索前列醇灌胃早孕Wistar大鼠制作药物流产模型,给予五加生化胶囊混悬液进行干预,观察大鼠子宫形态、子宫出血量及出血时间,以柱前衍生-气质联用技术检测大鼠血清代谢产物变化,运用主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)等技术对差异代谢产物进行分析,结合数据库揭示相应机制。结果:依据相对峰面积可知,与正常组比较,模型组大鼠血清中甘氨酸、丝氨酸、氨基丙二酸、L-焦谷氨酸、L-鸟氨酸、L-酪氨酸、花生四烯酸和胆固醇含量明显降低,2-丁烯二酸、磷酸甘油酯、L-赖氨酸、D-半乳糖、D-葡萄糖酸、2-单棕榈酸甘油酯和2-单硬脂酸甘油酯含量明显升高,给予五加生化胶囊混悬液后明显回调。结论:五加生化胶囊对早孕大鼠药物流产出血有一定治疗作用,其作用可能是通过调节脂肪酸代谢及氨基酸生成等途径实现的。Objective:To explore the effect and the mechanism of Wujia Shenghua Capsule in treating bleeding after medical abortion in early pregnant rats with the metabonomics method.Methods:Mifepristone and Misoprostol were given to early-pregnancy rats to establish the model of bleeding after medical abortion.For intervention,Wujia Shenghua Capsule suspension was given to the model rats.The histology of uterus,the amount of bleeding and the days of bleeding lasted were observed for examination.Pre-column derivatization-GC-MS was used to detect the changes of serum metabolites.PCA,OPLS-DA and other techniques were used to analyze the different metabolites,and the corresponding mechanism was revealed based on the database.Results:According to the relative peak areas,Compared to those in the normal group,the serum contents of glycine,serine,anilinomalonic acid,L-pyroglutamic acid,L-ornithine,L-tyrosine,arachidonic acid and cholesterol were significantly decreased,whereas the concentrations of L-fumaric acid,phosphoglyceride,L-lysine,L-galactose,D-gluconic acid,2-monopalmitin and 2-glyceryl monostearate were significantly increased in the model group.All the indicators were obviously reversed after the intervention of Wujia Shenghua Capsule.Conclusion:Wujia Shenghua Capsule has a good therapeutic effect on bleeding after medical abortion in early pregnant rats.Its mechanism may be achieved by regulating fatty acid metabolism and promoting amino acid production.
