复方栀子金花清热泻火药理作用研究

摘　　要：目的:研究复方栀子金花的清热泻火药理作用及其作用机制。方法:建立拘束应激大鼠模型和二甲苯诱导的小鼠耳肿胀模型,考察栀子金花复方对拘束应激大鼠和小鼠耳肿胀的治疗作用;称量大鼠胸腺及脾脏的重量并计算免疫器官指数,测定其血清丙氨酸氨基转移酶(ALT)、抗氧化能力(T-AOC)、甘油三酯(TG)、超氧化物歧化酶(SOD)、白介素-2(IL-2)、白介素-3(IL-3)及肝组织中丙二醛(MDA)、一氧化氮(NO)、黄嘌呤氧化酶(XOD)的含量和抗氧化能力指数(ORAC);称量小鼠耳片重量计算肿胀抑制率,测定其血清中肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)和白介素-8(IL-8)的含量。结果:复方栀子金花可以明显抑制拘束应激所致的免疫器官指数的降低,降低拘束应激大鼠血清ALT及肝组织中MDA的含量,显著提高其T-AOC指数和TG清除率;改善二甲苯诱导的小鼠耳肿胀度,显著降低血清中TNF-α、IL-1β、IL-6和IL-8的含量。结论:复方栀子金花具有较好的清热泻火作用,其作用机制可能与调节免疫力和降低炎症因子相关。Objective:To investigate the effect and the action mechanism of compound Zhizi Jinhua in terms of clearing heat and purging fire.Methods:The model of restraint stress depression was established in the rats,and the ear swelling model was established in the mice,thus to observe the influence of compound Zhizi Jinhua to restraint stress rats and ear swelling mice.The immune organ indexes were calculated by weighting the thymus and spleen.The serum levels of alanine aminotransferase(ALT),total antioxidant capacity(T-AOC),triglyceride(TG),superoxide dismutase(SOD),interleukin-2(IL-2)and interleukin-3(IL-3),the contents of malondialdehyde(MDA),nitric oxide(NO),xpurine oxidase(XOD)in liver tissues,as well as antioxidant capacity index(ORAC)were evaluated in the rats.The weight of the mice ear pieces was measured,the swelling inhibition rates were calculated,the serum contents of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6)and interleukin-8(IL-8)were detected in the mice.Results:It found that compound Zhizi Jinhua could inhibit the decreases of immune organ indexes caused by the restraint stress,lower the levels of ALT and MDA,and increase T-AOC index and TG clearance ratio significantly.Meanwhile,compound Zhizi Jinhua could improve the ear swelling induced by xylene in mice,significantly reduce the serum contents of TNF-α,IL-1β,IL-6 and IL-8.Conclusion:Compound Zhizi Jinhua has a good effect of clearing heat and purging fire,which mechanism may be related to the regulation of immunity and the reduction of inflammatory factors.
