摘 要:目的:观察过表达生存素(Survivin)对颅脑外伤大鼠神经细胞凋亡和神经功能的影响。方法:75只SD大鼠随机分为对照组、模型组和Survivin组。模型组和Survivin组大鼠采用自由落体法构建颅脑外伤大鼠模型,假手术组不做颅脑损伤。假手术组和模型组大鼠经颅内注射对照线相关病毒1×1011 vg,Survivin组大鼠经颅内注射携带Survivin基因的腺相关病毒1×1011 vg。治疗4周后,采用神经行为学评分评价3组大鼠神经功能状态,采用Morris水迷宫分析3组大鼠的学习记忆能力。采用原位缺口末端标记法(TUNEL)染色分析3组大鼠神经元凋亡水平。采用荧光定量聚合酶链反应分析Survivin、B细胞淋巴瘤/白血病-2(bcl-2)、bcl-2相关X蛋白(bax)和半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)mRNA表达水平。组间比较采用单因素方差分析。结果:Survivin组大鼠脑组织Survivin mRNA表达水平(3.17±0.32)明显高于假手术组(1.15±0.22)和模型组(1.10±0.18),差异有统计学意义(t=4.088、3.591,P<0.05)。Survivin组大鼠神经功能评分[(6.88±1.01)分]明显低于模型组大鼠神经功能评分[(10.79±1.54)分],差异有统计学意义(t=4.319,P<0.05)。Survivin组大鼠逃避潜伏期[(7.88±0.93)s]低于模型组大鼠逃避潜伏期[(10.98±2.21)s],差异有统计学意义(t=3.114,P<0.05)。Survivin组大鼠穿台指数(5.27±0.99)高于模型组大鼠穿台指数(3.01±0.89),差异有统计学意义(t=2.891,P<0.05)。Survivin组大鼠神经细胞凋亡比例[(14.21±3.25)%]明显低于模型组大鼠神经细胞凋亡水平[(25.91±5.20)%],差异有统计学意义(t=3.972,P<0.05)。Survivin组大鼠脑组织bax和Caspase-3 mRNA表达水平(1.79±0.24、1.44±0.17)明显低于模型组bax和Caspase-3蛋白表达水平(2.19±0.38、1.95±0.22)比较,差异有统计学意义(t=2.528、2.107,P<0.05)。Survivin组大鼠脑组织bcl-2 mRNA水平(0.91±0.13)明显高于模型组(0.49±0.11),差异有统计学意义(t=3.027,P<0.05)。结论:过表达SObjective To investigate the effect of overexpression of Survivin on neuronal apoptosis and neurological function in rats with traumatic brain injury.Methods Totally,75 SD rats were randomly divided into control group,model group and Survivin group.The rats in the model group and Survivin group were used to establish the rat model of craniocerebral injury by free fall,and the animals in the sham operation group were not subjected to craniocerebral injury.Rats in sham operation group and model group were injected with 1×1011 vg of control line associated virus,and rats in Survivin group were injected with 1×1011 vg of adeno-associated virus carrying Survivin gene.After 4 weeks of treatment,neurobehavioral score was used to evaluate the neurological function of the three groups.The learning and memory ability of the three groups was analyzed by Morris water maze.The level of neuronal apoptosis in the three groups was analyzed by terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL).The mRNA expression levels of Survivin,B cell lymphoma/leukemia-2(bcl-2),bcl-2 associated X protein(bax)and cysteinyl aspartate-specific protease(Caspase)-3 were analyzed by fluorescence quantitative polymerase chain reaction.Univariate analysis of variance was used for comparison between groups.Results The expression level of Survivin mRNA in Survivin group(3.17±0.32)was significantly higher than that in sham operation group(1.15±0.22)and model group(1.10±0.18,t=4.088,3.591,P<0.05).The neurological function score in Survivin group(6.88±1.01)was significantly lower than that in model group(10.79±1.54,t=4.319,P<0.05).The escape latency in Survivin group[(7.88±0.93)s]was significantly lower than that in model group[(10.98±2.21)s,t=3.114,P<0.05].The puncture index in Survivin group(5.27±0.99)was significantly higher than that in model group(3.01±0.89,t=2.891,P<0.05).The neuronal apoptosis rate in Survivin group[(14.21±3.25)%]was significantly lower than that in model group[(25.91±5.20)%,t=3.972,P<0.05].The exp
