摘 要:目的探讨阴虚动风证帕金森病(PD)病证一体动物模型小肠碳末推进率、胃窦连接蛋白(Cx43)的表达、血清中部分胃肠激素的变化及复方地黄颗粒的干预作用。方法 SD大鼠右侧黑质注射6-羟基多巴胺制备阴虚动风证PD大鼠模型,检测PD模型大鼠、复方地黄颗粒组大鼠小肠碳末推进率,免疫组化法检测胃窦部位Cx43的表达,酶联免疫吸附法(ELISA)检测血清中胃动素(MTL)、血管活性肠肽(VIP)、P物质(SP)水平。结果与正常对照组比较,PD模型组大鼠小肠碳末推进率减少,胃窦部Cx43数量减少,血清中MTL水平下降,SP、VIP水平升高,差异均具有统计学意义(P<0.05)。与模型组比较,复方地黄颗粒组大鼠小肠碳末推进率增加,胃窦部Cx43数量明显增加,血清中MTL水平上升,SP、VIP水平降低,差异均具有统计学意义(P<0.05),接近正常对照组水平。结论阴虚动风证帕金森病病证一体动物模型大鼠胃肠动力减低,与胃窦部连接蛋白Cx43的表达、血清中MTL、VIP、SP水平有关;中药复方地黄颗粒能有效增加胃肠动力,该作用与调节胃窦部Cx43的表达量、调节血清MTL、VIP、SP水平有关。Objective To investigate the small intestinal carbon advancing rate, the expression of gastric antrum connexin43(Cx43), the changes of some gastrointestinal hormones in serum, and the intervention effect of the compound Dihuang Granules in rats model of Parkinson’s disease(PD) with Yin deficiency and moving wind syndrome. Methods 6-Hydroxydopamine was injected into the substantia nigra of the right side of the rats to prepare a PD rat model with Yin deficiency and dynamic wind syndrome. The PD model rats and the compound Dihuang Granules group were tested for the small intestinal carbon powder advancing rate. The expression of Cx43 was detected by ELISA to detect the levels of motilin(MTL), vasoactive intestinal peptide(VIP), and substance P(SP) in serum.Results Compared with the normal control group, in the PD model group, the small intestinal carbon advancement rate was reduced, the amount of Cx43 in the antrum of the stomach was reduced, the serum MTL level decreased, and the SP and VIP levels increased(P<0.05). Compared with the model group, in the compound Dihuang Granules group, the intestinal carbon advancement rate of rats increased, the amount of Cx43 in the gastric antrum increased significantly, the serum MTL level increased, and the SP and VIP levels decreased(P<0.05), which were close to the normal control group. Conclusion The decrease of gastrointestinal motility in an animal model of Parkinson’s disease with syndrome of yin deficiency and movement of wind is related to the expression of connexin Cx43 in gastric antrum and the level of MTL, VIP and SP in serum;Chinese medicine compound Dihuang Granules can effectively increase the stomach intestinal motility, this effect is related to the regulation of the expression of Cx43 in the gastric antrum and the regulation of serum MTL, VIP, and SP levels.
