摘 要:目的探讨杞菊地黄丸方对妊娠期高血压大鼠可溶性血管内皮生长因子-1(sFIt-1)、胎盘生长因子(PLGF)水平及胎盘血管生成的影响。方法选取SD受孕大鼠30只,随机均分为对照组、模型组及杞菊地黄丸方组;模型组和杞菊地黄丸方组大鼠于受孕第13天皮下注射亚硝基左旋精氨酸甲酯[L-NAME,125 mg/(kg·d)、1次/d,共3 d],构建妊娠高血压模型,杞菊地黄丸方组大鼠于造模第3天给予杞菊地黄丸方灌胃[5 mL/(kg·d),2次/d,共5 d],对照组和模型组大鼠给予同等剂量的生理盐水灌胃;比较3组大鼠给药第10、15及19天时的血压和24 h尿蛋白水平,受孕第21天大鼠行剖宫产分娩,采用RT-PCR和Western blot法检测3组大鼠胎盘组织sFIt-1、PLGF、血管内皮生长因子(VEGF)mRNA和蛋白表达水平。结果妊娠第10天时,各组大鼠收缩压、舒张压及24 h尿蛋白水平比较,差异无统计学意义(P>0.05);妊娠第15天时,模型组和杞菊地黄丸方组大鼠收缩压、舒张压及24 h尿蛋白水平均较对照组明显升高(P<0.01);妊娠第19天时,杞菊地黄丸方组大鼠收缩压、舒张压和24 h尿蛋白均较模型组降低(P<0.05或P<0.01);妊娠第21天时,与对照组比较,模型组大鼠胎盘组织sFIt-1 mRNA和蛋白表达水平明显升高(P<0.01),PLGF和VEGF mRNA和蛋白表达水平明显降低(P<0.01);与模型组比较,杞菊地黄丸方组大鼠胎盘组织sFIt-1 mRNA和蛋白表达水平明显降低(P<0.01),PLGF和VEGF mRNA和蛋白表达水平明显升高(P<0.01)。结论杞菊地黄丸方可以降低妊娠期大鼠后期血压、24 h尿蛋白,机制可能与下调胎盘sFIt-1表达,上调PLGF和VEGF表达有关。Objective To investigate the effect of qijudihuang decoction on soluble vascular endothelial growth factor-1(sFIt-1),placental growth factor(PLGF)level and placental angiogenesis in rats with pregnancy-induced hypertension.Methods Thirty SD pregnant rats were randomly divided into control group,model group,and qijudihuang decoction group.Rats in model group and qijudihuang decoction group were injected L-NAME(125 mg/kg·d,once/d,three days in total)subcutaneously 13 days after conception to construct a model of pregnancy-induced hypertension,qijudihuang decoction group were given intragastrical qijudihuang decoction[5 mL/(kg·d)]twice/d,5 days in total,on the 3^(rd)day of modeling.Control group was injected subcutaneously with an equal amount of normal saline,and rats in the control group were given the same dose of intragastric normal saline.The changes of blood pressure and urine protein at 24 h in each group on 10^(th),15^(th),and 19^(th)day were observed.The c-section was performed on 21^(st) day after conception.RT-PCR and Western blot were used to detect the expression levels of sFIt-1,PLGF,VEGF mRNA and protein in the placenta tissue of each group.Results On the 10^(th) day of pregnancy,there were no significant differences in diastolic blood pressure,systolic blood pressure,and 24 h urine protein levels(P>0.05);on the 15^(th) day of pregnancy,the systolic blood pressure,diastolic blood pressure and 24 h urine protein levels of model group and the qijudihuang decoction group were significantly increased(P<0.01);on the 19^(th) day of pregnancy,the systolic blood pressure,diastolic blood pressure and 24 h urinary protein in the qijudihuang decoction group were significantly lower than those of model group(P<0.05 or P<0.01);on the 21^(st) day of pregnancy,compared with the control group,the expression levels of sFIt-1 mRNA and protein in placenta tissue of the model group rats were significantly increased(P<0.01),and the expression of PLGF and VEGF was significantly reduced(P<0.01).Compared with the model gro
