摘 要:目的:探讨藏医白脉疗法对局灶性脑缺血再灌注大鼠血液流变及炎症因子(IL-1β)的调节作用。方法:线栓法制作局灶性脑缺血再灌注大鼠模型。随机分为假手术组、模型组、白脉疗法组,脑缺血1.5 h,各组分别再灌注1 d、3 d。苏木精-伊红染色法(Hematoxylin-Eosinstaining,HE)染色观察皮层细胞形态变化,并应用血液流变技术检测血液流变情况,酶联免疫吸附试验ELISA和荧光定量PCR技术检测血中与大脑皮层炎症因子白介素1(IL-1β)的含量。结果:(1)HE染色表明白脉疗法组脑缺血大鼠皮层神经元的损伤程度低于模型组。(2)RTQ-PCR显示白脉疗法可以降低炎症因子(IL-1β)的表达,术后1 d、3 d炎症因子低于模型组,并且1 d具有显著性差异(P<0.05)。(3)酶联免疫吸附试验ELISA显示白脉疗法可以降低炎症因子(IL-1β)的表达,术后1 d、3 d炎症因子低于模型组且有显著性差异(P<0.05)。(4)血液流变表明3 d白脉疗法能够降低脑缺血大鼠血液粘度。结论:白脉疗法能够降低局灶性脑缺血再灌注大鼠血液粘度与炎症因子水平,对脑损伤有一定的保护作用。Objective:To investigate the effect of the Baimai(BM)therapy on blood rheology and Inflammatory factors(IL-1β)in rats with focal cerebral ischemia reperfusion.Methods:The rat model of focal cerebral isch⁃emia-reperfusion was established by suture method.They were randomly divided into sham operation group,model group and Baimai therapy group.Cerebral ischemia lasted for 1.5 hours and reperfusion lasted for 1 day and 3 days respectively.Morphological changes of cortical cells were observed by hematoxylin eosin staining(He⁃matoxylin-Eosin staining,HE),and hemorheology was used to detect hemorheology;The levels of interleukin 1(IL-1β)in the blood and cerebral cortex were detected by enzyme linked immunosorbent assay(ELISA)and Flu⁃orescence quantitative PCR.Results:(1)HE staining showed that the damage degree of cortical neurons in Bai⁃mai therapy group was lower than that in model group.(2)RTQ-PCR showed that Baimai(BM)therapy could re⁃duce the expression of inflammatory factors(IL-1β),and inflammatory factors in 1 days and 3 days after opera⁃tion were lower than those in the model group,and there was a significant difference in 1st day(P<0.05).(3)En⁃zyme linked immunosorbent assay(ELISA)showed that Baimai(BM)therapy could reduce the expression of in⁃flammatory factors(IL-1β),and inflammatory factors in 1 day and 3 days after operation were significantly lower than that in the model group(P<0.05).(4)Blood rheology showed that 3 days of Baimai(BM)therapy can reduce the blood viscosity in rats with cerebral ischemia.Conclusions:Baimai(BM)therapy can reduce the level of blood viscosity and inflammatory factors in rats with focal cerebral ischemia-reperfusion,and have certain pro⁃tective effects on brain injury.
