摘 要:为了选育出具有抗根肿病4号生理小种的甘蓝型油菜OguCMS恢复系,本研究以‘华抗5号’为供体,以甘蓝型萝卜质(OguCMS)恢复系RF04为受体亲本杂交,然后与RF04进行回交,构建培育新型恢复系选择群体,利用与萝卜质(OguCMS)恢复基因、抗根肿病基因紧密连锁的特异引物标记进行筛选,筛选出20个优良株系。通过实验室接种鉴定发现,RF04 100%感病且发病等级都为3级,而‘华抗5号’及F1代表现出100%抗病,发病等级为0级,说明F1代已经有抗病基因位点PbBa8.1;利用PbBa8.1基因紧密联锁标记(cnu_m090a, sau_um353a, A08-300)鉴定发现,BC2F2群体中均含有目标扩增片段,进一步说明基因片段PbBa8.1已转入回交群体;通过田间抗性鉴定,最终选择出2份株系在不同地点均表型高抗病,为C21906271-1和C21906145-1;1份材料(C21906273-2)在武汉表现出高抗病,在安徽表型抗病。本研究为利用分子标记辅助选择技术培育具有抗根肿病基因的甘蓝型春油菜提供了理论基础和技术途径。To breed a Brassica napus L.OguCMS restorer line(containing PbBa8.1 locus resistant to race 4 of plasmodiophora brassicae Woronin).This study used’Huakang5’as the donor and a cabbage radish(OguCMS)restorer line RF04 as the recipient parent Crossed and then backcrossed with RF04 to construct a breeding population for selection of new restorer lines.Screening was performed using specific primer markers that were closely linked to the radish quality(OguCMS)restorer gene and resistance to rhizome disease gene,and 20 excellent strains were obtained.Through laboratory vaccination identification,it was found that RF04 was 100%susceptible and the incidence grade was level 3,while’Huakang5’and F1 generations showed 100%disease resistance and the incidence grade was 0.It indicates that the F1 generation already has the disease resistance gene locus PbBa8.1,the use of the PbBa8.1 gene tightly linked markers(cnu_m090 a,sau_um353 a,A08-300)identified that the target BC2 F2 population contained the amplified fragment,further explaining the site PbBa8.1 has been transferred to backcross populations.Through field resistance identification,2 strains were selected at the same time and all had high phenotypic resistance at different locations,which were C21906271-1 and C21906145-1,1 material(C2190-6273-2)high resistance in Wuhan and phenotypic resistance in Anhui.This study provides a theoretical basis and technical approach for the cultivation of spring rapeseed(Brassica napus L.)with a gene for resistance to rhizobia using molecular marker-assisted selection technology.
