摘 要:为短期内获得大量优质的粉葛丛生芽,从而筛选粉葛丛生芽诱导的适宜条件。本研究以粉葛(Pueraria montana var. thomsonii)幼嫩带节茎段为外植体,探索外植体消毒条件,腋芽萌发培养基、丛生芽诱导培养基丛生芽增殖培养基。结果表明:(1)外植体最佳消毒条件为75%酒精浸泡30 s后,0.1%氯化汞溶液(加2滴吐温80℃)处理10 min,污染率为10.00%,腋芽萌发率为93.33%。(2)最佳腋芽生长培养基为MS+6-BA 0.2 mg/L+NAA 0.1 mg/L+活性炭0.2 g/L,腋芽萌发率达93.33%。(3) MS+TDZ1.5 mg/L+NAA 0.1 mg/L+酸水解酪蛋白200 mg/L为最佳丛生芽诱导培养基,丛生芽诱导率达90.00%。(4) MS+IBA 0.2 mg/L+6-BA 1.0 mg/L+KT 1.0 mg/L为最佳丛生芽增殖培养基,增殖系数为1.77。本研究结果可为基于丛生芽诱导的粉葛离体快繁体系建立提供依据。In order to obtain a large number of high-quality cluster buds of Pueraria montana var.thomsonii in a short period of time,suitable conditions for induction of cluster buds of Pueraria montana var.thomsonii were screened.In this study,the young stems of Pueraria montana var.thomsonii with axillary buds were used as explants,culture medium for germination of axillary buds,induction medium and proliferation mediums for cluster buds were explored.The results were as follows:(1)The best disinfection conditions for explants was soaked with75%alcohol for 30 s,followed by treatment with 0.1%mercuric chloride solution(Including 2 drops of Twain 80℃)for 10 min.The contamination rate was 10.00%and germination rate was 93.33%.(2)The optimum medium for germination of axillary buds was MS+6-BA 0.2 mg/L+NAA 0.1 mg/L+activated carbon 0.2 g/L,and the axillary buds germination rate reached 93.33%.(3)The best induction medium for cluster buds was MS+TDZ 1.5 mg/L+NAA0.1 mg/L+acid-hydrolyzed casein 200 mg/L,and the induction rate reached 90.00%.(4)The best proliferation medium for the cluster buds was MS+IBA 0.2 mg/L+6-BA 1.0 mg/L+KT 1.0 mg/L,and the proliferation coefficient was 1.77.The results can provide t heoretical foundation for the establishment of an in vitro rapid propagation system which is based on tufted bud induction of Pueraria montana var.thomsonii.
