摘  要:目的通过网络药理学探讨黄芪、附子治疗重症肌无力(myasthenia gravis,MG)的药理机制,为益气温阳法的临床应用提供理论依据。方法总结归纳出用益气温阳法治疗MG患者的主要中医临床症状,利用TCMSP、Swiss Target Prediction预测和筛选黄芪、附子的活性成分与作用靶点,用Gene Cards分别收集症状和MG疾病相关靶点,并在Venny 2.1. 0获取共同靶点,通过R语言进行GO和KEGG富集分析,预测其作用机制。结果筛选出38个化学成分和796个靶点,83个共同靶点,包括AKT1、IL6、TNF、VEGFA、TP53、JUN、MAPK1等。黄芪、附子治疗重症肌无力机制可能涉及病毒感染通路、免疫相关通路、PI3K-Akt信号通路等。结论黄芪、附子治疗MG的药理机制是通过多组分、多靶点、多通路实现的,用益气温阳法治疗MG有一定的临床价值。Objective To explore the pharmacological mechanism of Astragali and Aconite treatment of myasthenia gravisthrough network pharmacology and provide theoretical basis for the clinical application of benefiting qi and warming yang Methods.Methodssummarizing the main clinical symptoms of traditional Chinese medicine in myasthenia gravis patients treatedwith benefiting qi and warming yang method. Using TCMSP and Swiss Target Prediction to predict and screen the active com-ponents and targets of Astragalus and Aconite. Using Gene Cards to collect symptoms and MG-related targets, and obtaincommon targets for in Venny 2. 1. 0. Enrichment analysis of GO and KEGG through R language, predict its mechanism of action.ResultsScreened 38 chemical components and 796 targets of Astragalus and Aconite, and 83 common targets,includingAKT1,IL6,TNF,VEGFA,TP53,JUN and MAPK1. The mechanism of Astragalus and Aconite treatment of myasthenia gravismay involve viral infection pathway, immune-related pathway and PI3 K-Akt signaling pathway.ConclusionThe pharmacological mechanism of Astragalus and Aconite treatment of MG achieved through multiple components, multiple targets, andmultiple pathways. The treatment of MG with benefiting qi and warming yang method has certain clinical value.
