摘 要:目的:探讨生酮饮食对小鼠骨骼肌力量及运动耐力的影响。方法:20只小鼠随机分为标准饮食组(n=10)和生酮饮食组(n=10),按等能量喂养原则,分别给予两组小鼠标准饲料和生酮饲料4周,每天记录体重和进食量。4周后通过尾静脉采血测定血糖、血酮水平;采用负重游泳实验和在体腓肠肌收缩实验评价小鼠运动能力和骨骼肌力量,并测试腓肠肌质量及总蛋白含量的变化。结果:4周后,生酮饮食组小鼠体重显著低于标准饮食组(P<0.05),而血酮水平显著高于标准饮食组(P<0.05)。小鼠负重游泳时间、腓肠肌单收缩最大幅度和强直收缩最大幅度、连续串刺激至最大收缩幅度后下降至最大幅度50%所用时间(T50)两组均无显著性差异(P>0.05)。两组小鼠腓肠肌的质量和总蛋白含量亦无显著性差异(P>0.05)。结论:为期4周的生酮饮食在有效降低体重的同时,对小鼠的骨骼肌力量和运动耐力无显著影响。Objective To explore the effects of ketogenic diet on skeletal muscle strength and athletic endurance of mice. Methods A total of 20 male ICR mice were randomly divided into a standard diet group(SD group) and a ketogenic diet group(KD group),each of 10. According to the principle of equal energy feeding,both groups were given standard and ketogenic food respectively for 4 weeks,and their body weight and food intake were recorded every day. Then blood samples were collected from the tail vein to measure the blood glucose and ketone levels. The athletic endurance and the strength of skeletal muscle were evaluated respectively using the weight-bearing swimming experiment and the gastrocnemius contraction experiment in vivo. And the mass and total protein content of gastrocnemius were compared between the two groups. Results After 4 weeks,the average body weight of the KD group was significantly lower than the SD group(P<0.05),while the average blood ketone level was significantly higher than the latter(P<0.05). No significant difference was observed in the average endurance of the weight-bearing swimming,the maximum amplitude of the single contraction and tetanic contraction of gastrocnemius,and the time of decreasing to 50% of maximal contraction after stimulating continuously to the maximum contraction amplitude(P>0.05). There was no significant difference in the average mass and total protein content of gastrocnemius muscle between the two groups(P>0.05). Conclusion Going on the four-week ketogenic diet can reduce the body weight of mice effectively,but have no effect on the skeletal muscle strength and athletic endurance.
