摘  要:目的:采用网络药理学的方法探讨香附治疗肝癌的作用机制。方法:运用中药系统药理学数据库与分析平台检索香附的活性成分及其作用靶点,通过GeneCards和OMIM数据库检索肝癌的相关靶点;应用Cytoscape 3.7.0软件构建“香附-活性成分-肝癌-靶点”相互作用网络,通过STRING数据库分析绘制靶点与蛋白相互作用的PPI可视化网络图。运用R语言软件对药物作用靶点与疾病靶点的交集进行核心靶基因的GO与KEGG通路的富集分析。结果:检索筛选得到香附的有效活性成分16个,主要包括异鼠李素、β-谷甾醇、木樨草素、槲皮素和8-异戊烯基-山奈酚等。PPI网络包含105个节点,937条边。GO富集分析涉及的生物学功能包括DNA结合转录激活剂活性、DNA结合转录因子结合、神经递质结合等。KEGG所涉及的生物学通路及生物学过程包括PI3K-Akt信号通路、Kaposi肉瘤相关疱疹病毒感染、流体剪切应力与动脉粥样硬化及细胞凋亡信号通路等。结论:香附中的有效活性成分通过多个靶点及通路治疗肝癌,为进一步研究香附的抗肿瘤作用机制提供了理论依据。Objective:To inquire into the mechanism of Xiangfu(Rhizoma Cyperi) in treating liver cancer based on network pharmacology.Methods:The active ingredients and their targets of Xiangfu were searched by the pharmacology database and analysis platform of TCM system,and the related targets of liver cancer were searched by GeneCards and OMIM database.Cyposcape 3.7.0 software was used to construct the interaction network of "Xiangfu-active ingredients-liver cancer-target",and the PPI visualization network diagram of the interaction between target and protein was drawn by STRING database analysis.R language software was used for enrichment analysis of GO and KEGG pathways of core target genes of intersection of drug targets and disease targets.Results:A total of 16 active components of Xiangfu were screened,mainly including isorhamnetin,β-sitosterol,luteolin,quercetin and 8-isopentenyl-kaempferol etc.PPI network contained 105 nodes and 937 edges.The biological functions of GO enrichment analysis included DNA binding transcriptional activator activity,DNA binding transcription factor connection,neurotransmitter binding,etc.The biological pathway and biological process involved in KEGG included PI3 K-Akt signaling pathway,Kaposi sarcoma related herpes virus infection,fluid shear stress,atherosclerosis and apoptosis signaling pathway,etc.Conclusion:The active components in Xiangfu can treat liver cancer through multiple targets and pathways,which provides a theoretical basis for further study of the anti-tumor mechanism of Xiangfu.
