摘  要:目的研究“人参-鹿茸”治疗阿尔茨海默症的主要药效物质基础及作用机制。方法利用TCMSP和TCM筛选并建立“人参-鹿茸”有效成分数据库;通过Swiss Target Prediction和TTD数据库分别预测有效成分和阿尔茨海默症的相关靶点并使其映射;对映射所得靶点进行GO功能分析和KEGG信号通路分析;利用Cytoscape(3.7.1)软件构建“成分-靶点-通路”网络;根据贡献指数分析确定“人参-鹿茸”治疗阿尔茨海默症的物质基础。结果“人参-鹿茸”共筛选出29个重要潜在活性成分,与其相关的阿尔茨海默症的靶点40个,9条KEGG信号通路和12条生物过程注释。结论“人参-鹿茸”药对以Retinol、17-Beta-Estradiol、Panaxadiol、Ginsenoside Rb1、Kaempferol和Adenosine Triphosphate构成主要物质基础,通过调节PTGS2、DRD2、ACHE、BACE1和TRPV1等靶点并作用到神经活性配体-受体相互作用、血清素能突触、cAMP信号通路、PI3K-Akt信号通路和癌症途径等信号通路中,进而发挥治疗阿尔茨海默症的作用。本研究揭示了“人参-鹿茸”通过“多成分-多靶点-多途径”的方式调控阿尔茨海默症的主要物质基础和作用机制,为进一步开发“人参-鹿茸”提供了理论基础和科学依据。Objective To study the main pharmacodynamic material basis and mechanism of the couplet medicines of ginseng and pilose antler in the treatment of Alzheimer’s disease.Methods TCMSP and TCM were used to sort out the active components of ginseng and pilose antler and establish its database.Swiss Target Prediction and TTD databases were respectively used to predict and map the active components and the related targets of Alzheimer’s disease.GO function analysis and KEGG signaling pathway analysis were performed on the mapped targets.Cytoscape(3.7.1)software was used to construct a“component-target-pathway”network.The material basis of the couplet medicines of ginseng and pilose antler was determined in the treatment of Alzheimer’s disease according to the contribution index analysis.Results A total of 29 important potential active components were screened out from the couplet medicines of ginseng and pilose antler,and 40 targets of Alzheimer’s disease,9 KEGG signaling pathways and 12 bioprocess annotations were proved to be related to those active components.Conclusion The couplet medicines of ginseng and pilose antler take Retinol,17-Beta-Estradiol,Panaxadiol,Ginsenoside Rb1,Kaempferol and Adenosine Triphosphate as their main material basis.And by regulating targets such as PTGS2,DRD2,ACHE,BACE1 and TRPV1,the active components act on the neuroactive ligand-receptor interactions,serotonergic synapses,cAMP signaling pathways,PI3K-Akt signaling pathways and cancer pathways and other signaling pathways,so as to play a role in the treatment of Alzheimer’s disease.This study reveals the main material basis and action mechanism of the couplet medicines in regulating Alzheimer’s disease through the way of multi-component,multi-target and multi-pathway,and it provides a theoretical and scientifi c basis for the further development of the couplet medicines of ginseng and pilose antler.
