摘 要:目的分析八珍益智方中非挥发性成分在大鼠体内的代谢产物。方法大鼠灌胃给予八珍益智方水煎液(13.95 g/kg)后,采用超高效液相色谱⁃线性离子阱⁃静电轨道阱串联质谱联用(UHPLC⁃LTQ⁃Orbitrap⁃MS)法分析其血浆、胆汁、尿液、粪便中的非挥发性成分。结果共鉴定出6个原型及40个代谢产物,主要发生Ⅰ相代谢反应(氧化、去甲基化等)、II相代谢反应(磺化、葡萄糖醛酸化、甘氨酸或谷胱甘肽结合等)。结论该方法快速有效,可用于鉴定大鼠体内八珍益智方代谢产物,并为进一步研究该复方制剂的功效物质基础和药动学提供依据。AIM To analyze the metabolites of non⁃volatile components from Bazhen Yizhi Prescription in rats in vivo.METHODS Rats were given intragastric administration of water decoction of Bazhen Yizhi Prescription(13.95 g/kg),after which ultra⁃high performance liquid chromatography⁃linear ion trap⁃electrostatic orbit trap tandem mass spectrometry(UHPLC⁃LTQ⁃Orbitrap⁃MS)method was adopted in the analysis of non⁃volatile components in the plasma,bile,urine and fece.RESULTS Total six prototype components and forty metabolites were identified,which mainly occurred PhaseⅠmetabolic reactions(oxidation,demethylation,etc.)and Phase II metabolic reactions(sulfonation,glucuronidation,glycine or glutathione binding,etc.).CONCLUSION This rapid and effective method can be used for the identification of metabolites of Bazhen Yizhi Prescription in rats in vivo,and provide a basis for the further research of effective material basis and pharmacokinetics of this compound preparation.
