摘 要:目的研究天花粉Trichosanthes kirilowii Maxim的化学成分及其对谷氨酸损伤PC12细胞的保护作用。方法天花粉甲醇提取物的乙酸乙酯部位采用硅胶、MCI、反相半制备HPLC等进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用MTT法检测其细胞活力,LDH法检测乳酸脱氢酶的漏出率。结果从中分离得到9个化合物,分别鉴定为大豆脑苷Ⅰ(1)、(24ξ)⁃24⁃乙基⁃胆甾⁃3⁃O⁃β⁃D⁃葡萄糖苷(2)、(24ξ)⁃24⁃乙基⁃胆甾⁃22⁃烯⁃3⁃O⁃β⁃D⁃葡萄糖(3)、ligballinol(4)、亚油酸甘油酯(5)、亚油酸(6)、(9Z,12Z)⁃9,12⁃十九碳二烯酸(7)、正十六烷酸(8)、正二十二烷酸(9)。化合物1能明显改善PC12细胞被谷氨酸损伤的形态,细胞存活率显著升高,LDH释放减少。结论化合物2~3、5、7为首次从栝楼属植物中分离得到,化合物6、8~9为首次从该植物中分离得到。其中化合物3为不常见甾体糖苷,首次报道其NMR数据。化合物1可减轻谷氨酸诱导的PC12细胞损伤。AIM To study the chemical constituents from Trichosanthes kirilowii Maxim and their protective effects on glutamate⁃induced excitotoxicity in PC12 cells.METHODS The ethyl acetate fraction of methanol extract from T.kirilowii was isolated and purified by silica,MCI and semi⁃preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.MTT and LDH assays were applied to evaluating the cell viability and LDH leakage,respectively.RESULTS Nine compounds were isolated and identified as soya⁃cerebroside I(1),(24ξ)⁃24⁃ethyl⁃cholesta⁃3⁃O⁃β⁃D⁃glucoside(2),(24ξ)⁃24⁃ethyl⁃cholesta⁃22⁃en⁃3⁃O⁃β⁃D⁃glucoside(3),ligballinol(4),1⁃O⁃linoleoyl⁃glycerol(5),linoleic acid(6),(9Z,12Z)⁃9,12⁃nonadecadienoic acid(7),hexadecanoic acid(8),and behenic acid(9).Compound 1 could significantly improve the morphology of PC12 cells,increase the cell viability and decrease leakage of LDH.CONCLUSION Compounds 2-3,5,7 are first isolated from the genus Trichosanthes,and compounds 6,8-9 are isolated from this plant for the first time.Compound 3 is a less common sterol glucoside,and its NMR data are first reported.Compound 1 can obviously alleviate glutamate⁃induced injury in PC12 cells.
