摘 要:目的探讨益气活血方是否通过调节自噬相关基因Beclin1和Atg5的表达调控发挥对心肌缺血的保护作用。方法将60只健康SD大鼠随机分成空白组、模型组、地奥心血康组、益气活血方高、中、低剂量组,通过腹腔注射异丙肾上腺素制备急性心肌缺血损伤模型。观察心肌梗死面积、心肌细胞超微结构,检测自噬基因Atg5、Beclin1 mRNA和凋亡基因Caspase-3 mRNA的表达。结果益气活血方高、中剂量组各项指标均明显高于模型组,益气活血方各剂量组大鼠心肌组织中Atg5、Beclinl mRNA表达量均升高(P<0.01或P<0.05),Caspase-3mRNA表达量降低(P<0.01或P<0.05)。结论益气活血方可以通过上调自噬相关基因Atg5、Beclin1 mRNA表达,增强细胞自噬,下调Caspase-3mRNA表达,从而发挥对心肌缺血的保护作用。Objective To investigate whether Yiqi Huoxue Recipe exerts a protective effect on myocardial ischemia by regulating the expression of autophagy-related genes Beclin1 and Atg5.Methods60 healthy SD rats are randomly divided into a blank group,a model group,a Diaoxinxuekang group,and a high,medium and low dose of Yiqi Huoxue Recipe.The acute myocardial ischemic injury model is prepared by intraperitoneal injection of isoproterenol.Observe the area of myocardial infarction,the ultrastructure of myocardial cells,and detect the expression of autophagy gene Atg5,Beclin1 mRNA and apoptosis gene Caspase-3 mRNA.Results The indexes of Yiqi Huoxue Recipe high and medium dose groups are significantly higher than those of the model group.The expression of Atg5 and Beclinl mRNA in rat myocardial tissues in each dose group of Yiqi Huoxue Recipe is significantly up-regulated(P<0.01 or P<0.05),Caspase-3 mRNA expression is significantly down-regulated(P<0.01 or P<0.05).Conclusion Yiqi Huoxue Recipe can up-regulate autophagy-related genes Atg5 and Beclin1 mRNA expression,enhance cell autophagy,and down-regulate Caspase-3 mRNA expression,thereby exerting a protective effect on myocardial ischemia.
