摘  要:目的评估苦参中的4种主要生物碱与洗必泰对变形链球菌的浮游细胞代谢活性以及生物被膜形成的协同抑制作用。方法通过MTT法测定了4种生物碱对变形链球菌浮游细胞代谢活性的影响,通过结晶紫染色法测定了4种生物碱对生物被膜形成的抑制作用,通过棋盘法以及零相互作用效能模型(ZIP)分析了4种生物碱与洗必泰对变形链球菌的协同抑制作用。结果4种生物碱单独处理时对变形链球菌浮游细胞的代谢活性以及生物被膜的形成都表现出浓度依赖性的抑制作用,与洗必泰联合使用时也对变形链球菌具有不同程度的协同抑制作用,其中苦参碱与洗必泰的协同活性最强。结论苦参中的4种主要生物碱增强了洗必泰对变形链球菌浮游细胞和生物被膜细胞的体外抗菌活性,为天然药物与传统抗菌药物的联用提供了新思路。Objective To evaluate the synergistic inhibitory effects of the four major alkaloids in Sophora flavescens with chlorhexidine on the planktonic cell metabolism and biofilm formation of Streptococcus mutans.Methods The effects of the four alkaloids on the metabolic activity of S.mutans planktonic cells are determined by MTT method,and the inhibitory effects of the four alkaloids on the biofilm formation are determined by crystal violet staining.The checkerboard method and Zero Interaction Potency model(ZIP)analyzed the synergistic inhibitory effects of the four alkaloids with chlorhexidine on S.mutans.Results The four alkaloids exhibited a does-dependent inhibitory effect on the metabolic activity of plankton cells and the biofilm formation of S.mutans after individual treatment.When used in combination with chlorhexidine,they also has different degrees of synergistic inhibitory effects on S.mutans.Among them,matrine have the strongest synergistic activity with chlorhexidine.Conclusion The four alkaloids in Sophora flavescens enhance the antibacterial activity of chlorhexidine against S.mutans planktonic cells and biofilm cells,providing new light on the potential to develop natural medicine in combination with traditional antibacterial agents as treatments to arrest dental caries.
