宁夏民间药鸡娃草的质量标准研究

摘　　要：目的对鸡娃草药材质量标准进行研究。方法对鸡娃草药材进行性状、显微及TLC定性鉴别;根据药典通则测定鸡娃草的水分、灰分、醇溶性浸出物含量;利用HPLC法测定鸡娃草全草及不同颜色不同药用部位中白花丹素的含量。结果药材显微特征显著,薄层色谱图斑点清晰,分离度好;初步拟定宁夏地区鸡娃草药材水分不得超过6.0%;总灰分不得超过15.0%;酸不溶性灰分不得超过8.0%;醇溶性浸出物不得少于2.0%;白花丹素进样量在0.0290~0.2900μg/mL范围内呈良好的线性关系;加样回收率均值为99.82%,RSD为1.97%,10批鸡娃草药材中白花丹素的量为0.078%~0.160%,发现鸡娃草药材不同颜色根、茎、叶中白花丹素的含量差异较为明显,绿色根和红色叶中的白花丹素含量相对较高,分别为0.344%和0.321%。结论建立的鸡娃草药材质量标准研究方法可行,重复性好,可为宁夏民间药鸡娃草质量标准的制定奠定基础,也为其合理开发应用提供依据。Objective To establish quality standards for Plumbagella micrantha.Methods Morphological identification,microscopic method and TLC method are employed for P.micrantha identification.The contents of water,ash,alcohol-soluble extract of P.micrantha are determined with legal principles.The plumbagin content of P.micrantha is determined by HPLC.Results Microscopic features of P.micrantha are distinctive,and the spots of plumbagin in TLC are clear with better resolution.Moisture is not more than 6.0%,total ash is not more than 15.0%,acid insoluble ash is not more than 8.0%and alcohol-soluble extract is not less than 2.0%of folk medicine P.micrantha in Ningxia are studied out elementarily.The results of HPLC show that plumbagin displayed good linearity within the range of 0.0290~0.2900μg/mL.The average recoveries of plumbagin are 99.82%,with RSD of 1.97%.Plumbagin content of ten batches of P.micrantha in Ningxia within 0.078%~0.160%.We also find that there are significant differences on the content of plumbagin in the roots,stems and leaves of different colors of P.micrantha,the content of plumbagin in the green roots is 0.344%and red leaves is 0.321%,relatively higher than others.Conclusion This study on quality standards for folk medicine P.micrantha in Ningxia is feasible with good repeatability which can lay the foundation for the establishment of quality standard and provide reference for the development and rational utilization of the folk medicine.
