摘 要:目的探讨miR-29a在急性肾损伤(AKI)中的作用。方法 12只雄性CD1小鼠随机分为2组:对照组、叶酸组。检测血清肌酐、尿素氮,PAS染色观察病理变化,TUNEL染色观察细胞凋亡程度,qPCR检测基因IL-6、TNF-α、α-SMA、TGF-β1、miR-29a的表达,Western blot检测α-SMA、TGF-β1蛋白的表达。结果与对照组比,叶酸组血清肌酐、尿素氮升高,PAS染色显示肾小管扩张、刷状缘消失,TUNEL染色提示细胞凋亡增加,IL-6、TNF-α、α-SMA、TGF-β1表达上调,miR-29a表达下调。结论 miR-29可能参与AKI的发生发展。Objective To investigate the role of miR-29 a in acute kidney injury(AKI). Methods Twelve male CD1 mice were randomly divided into: control group and the folic acid group. The serum creatinine and blood urea nitrogen were measured, pathological changes were observed by PAS staining, apoptosis degree was observed with TUNEL staining, the expression of genes IL-6、 TNF-α 、 α-SMA、 TGF-β1 and miR-29 a were measured by qPCR. The expression of α-SMA, TGF-β1 protein were measured by Western blot. Results The contents of serum creatinine and blood urea nitrogen increased, dilation of renal tubules and disappearance of the brush border, apoptosis rate increased, the expression levels of IL-6 and TNF-α and α-SMA and TGF-β1 increased, the expression level of miR-29 a downregulated in folic acid group than in control group. Conclusion miR-29 a might be involved in the development of AKI.
