益气化瘀清热方及其拆方对IgA肾病大鼠蛋白尿、血尿及其血生化的影响

摘　　要：目的:探讨益气化瘀清热方及其拆方对IgA肾病(IgAN)大鼠蛋白尿、血尿及其血生化的影响。方法:将165只雄性Wistar大鼠随机分为11组,采用BSA+CCL4+LPS的实验方法制备IgAN模型,益气化瘀清热方干预;观察各组大鼠尿红细胞计数、24 h尿蛋白定量(UTP)、血生化情况。结果:在尿红细胞计数、24 h UTP方面,模型组第6周末时开始出现升高,并且逐渐加重。在第10周时,雷公藤多苷组、复方高剂量组与模型组比较明显下降(P<0.01);其余各组仍持续升高。第12周末时,各治疗组与模型组比较均显著降低(P<0.01)。复方高剂量组降低最明显,益气组最差;化瘀组和清热组表达高于复方组,低于益气组;化瘀高剂量组和清热高剂量组表达高于复方高剂量组,低于益气高剂量组;各组药物高剂量组表达低于低剂量组。在血生化结果方面,于第12周末检测实验大鼠血生化指标情况,各干预组大鼠TP、TCH、TG、Cr、BUN与空白组比较差异无统计学意义(均P>0.05);在ALT、AST方面,雷公藤多苷组与其他各组之间比较显著升高,差异有统计学意义(P<0.01);其他各组间比较无统计学意义。结论:益气化瘀清热方及其拆方能够较明显降低实验中IgAN大鼠血尿及蛋白尿情况,且对肝、肾功能无影响。Objective:To explore the effect of Yiqi Huayu Qingre Formula and its compositions on proteinuria,haematuria and blood biochemistry in rats with IgA nephropathy(IgAN).Methods:165 male Wistar rats were randomly divided into 11 groups.The experimental method of BSA+CCL4+LPS was applied to establish IgAN model,which was intervened with Yiqi Huayu Qingre Formula.The urinary red blood cell count,24-hour urine total protein(UTP)and blood biochemistry were observed in each group.Results:In terms of the urinary erythrocyte count and 24-hours UTP,they were started to rise from the end of the 6 th week and gradually aggravated;they were significantly decreased in the Tripterygium glycosides group and in the high-dose of compound group compared to those in the model group during the 10 th week(P<0.01),and they remained elevated in the rest groups;at the end of the 12 th week,they were decreased significantly in all medication groups compared to those in the model group(P<0.01),the decreases in the high-dose of compound group were most significant,whereas the decreases in the Yiqi group were the least significant.The Huayu group and the Qingre group were higher than the compound group,but lower than the Yiqi group;the high-dose of Huayu group and the high-dose of Qingre group were higher than the high-dose of compound group,but lower than the high-dose Yiqi group.Within the same medication groups,the high-dose group was better than the low-dose group.In terms of blood biochemistry,there were no statistical differences in TP,TCH,TG,Cr and BUN between the medication groups and the blank group at the end of the 12 th week(P>0.05).The levels of ALT and AST were significantly increased in the Tripterygium glycosides group compared to those in the other groups(P<0.01);whereas there were no statistical differences among the other groups.Conclusion:Yiqi Huayu Qingre Formula and its compositions can substantially improve the hematuria and proteinuria of the IgAN rats,and it had no adverse reactions to the functions of liver and kidney
