摘  要:目的通过HPLC测定山西产柴胡药材中柴胡皂苷a、d的含量,建立柴胡药材的HPLC指纹图谱,分析不同基源柴胡药材的质量差异,为山西省柴胡药材质量控制和规范化种植提供技术支持。方法采用《中华人民共和国药典》2020年版柴胡项下柴胡皂苷a、d含量测定的方法对不同基源的柴胡药材进行含量测定,建立柴胡药材的HPLC指纹图谱,利用相似度评价软件和多元统计分析对18批柴胡药材色谱图进行比较。结果不同基源柴胡中柴胡皂苷含量存在明显差异,山西产黑柴胡的柴胡皂苷含量低于北柴胡;北柴胡药材相似度较高,两种黑柴胡相似度也较高,但黑柴胡和北柴胡之间相似度相差较大;多元统计分析结果表明,三种不同基源的柴胡之间均有较大的差异,可以较好分开。结论山西产黑柴胡与北柴胡相比有较大的差异,所建立的HPLC指纹图谱能客观有效地评价不同产地柴胡药材的质量,可作为柴胡药材的质量控制有利手段。Objective To establish a HPLC fingerprint method for evaluating the quality of Bupleurum chinense DC.from different sources by determining the content of saikosaponin-a and saikosaponin-d,which can provide technical support to the quality control and standardized cultivation of Bupleuri Radix in Shanxi.Methods According to the current statutory standard method under Bupleurum chinense DC.text in the Chinese Pharmacopoeia(2020 edition)VolunmⅠ,the content of saikosaponin-a,-d were determined.In addition,HPLC fingerprint were established,and 18 sources Bupleuri Radix were compared by similarity evaluation software and multivariable statistics.Results The contents of saikosaponins show significant differences among different origin Bupleuri Radix and the saikosaponins in B.smithii from Shanxi were lower than those in B.chinense.B.smithii and B.chinense shared a high similarity,respectively,whereas those two origin plants varied widely.Three origin Bupleuri Radix could be distinguished easily by the analysis results of metabonomic.Conclusion The content of saikosaponins between B.smithii and B.chinense from Shanxi showed large difference.The established HPLC fingerprint is able to be used to assess the quality of different origin Bupleuri Radix efficiently and objectively,and it will be a favourable mean to control the quality of Bupleuri Radix.
