摘 要:目的采用复合磷脂脂质体技术以氢化大豆磷脂(HSPC)和大豆磷脂(SPC)为膜材制备丹参总酚酸复合磷脂脂质体,并研究其最佳制备工艺。方法采用逆向蒸发法制备复合磷脂脂质体,测定其粒径与粒径分布,以脂质体的成型性与包封率等为评价指标,研究丹参总酚酸脂质体的制备工艺。结果磷脂与胆固醇质量比为3∶1、有机相与水相体积比为4∶1、药脂比为1∶15、磷脂质量浓度为12.5 mg·mL^(-1)时得到的丹参酚酸B与丹参总酚酸的平均包封率分别为60.08%和69.45%,其中载药量以丹参总酚酸计达2.92%。结论该复合磷脂脂质体制备方法准确可靠,工艺合理稳定。Objective To prepare total salvianolic acid composite phospholipid liposome with hydrogenated soybean phosphatidylcholine and soybean phosphatidylcholine as the membrane materials by composite phospholipid liposome technology.Methods The composite phospholipid liposomes were prepared by reverse evaporation method.Particle size and distribution of the composite phospholipid liposomes were determined by nano-particle size measurement instrument.The preparation technology of total salvianolic acid composite phospholipid liposome was evaluated with the forming property and encapsulation rate of the liposomes.Results The average encapsulation rates of total salvianolic acid B and total salvianolic acid were 60.08%and 69.45%;the drug loading was 2.92%with total;while the mass ratio of phospholipid to cholesterol was 3∶1,the volume ratio of organic phase to aqueous phase was 4∶1,the drug lipid ratio was 1∶15,and the concentration of phospholipid was 12.5 mg·mL^(-1).Conclusion The preparation of composite phospholipid liposome is accurate and reliable and the technology is stable and reasonable.
