摘　　要：目的从抗炎、消肿及促进溃疡面愈合三个方面,对苦柏坐浴颗粒进行药效学评价,为苦柏坐浴颗粒的临床应用提供实验室依据。方法36只昆明种小鼠,随机分为空白对照组、马应龙麝香痔疮膏组、苦柏坐浴颗粒组,每组12只,分别于耳廓涂凡士林、马应龙麝香痔疮膏、苦柏坐浴颗粒,做小鼠耳肿胀实验。SD大鼠36只,造模成功后,随机分为模型对照组、马应龙麝香痔疮膏组、苦柏坐浴颗粒组,每组12只。分别在6、12、24、48、72 h于肛门肿胀处涂0.5%羧甲基纤维素钠(CMC-Na)溶液、马应龙麝香痔疮膏、苦柏坐浴颗粒,做肛周肿胀实验。SD大鼠36只,致溃疡后,随机分为模型对照组、马应龙麝香痔疮膏组、苦柏坐浴颗粒组,每组12只。分别给予凡士林基质、马应龙麝香痔疮膏、苦柏坐浴颗粒,做肛周溃疡面愈合实验。观察苦柏坐浴颗粒对小鼠耳肿胀的抗炎作用、对大鼠肛周肿胀的消肿作用、对大鼠肛周溃疡面愈合的影响。结果马应龙麝香痔疮膏组、苦柏坐浴颗粒组肿胀度分别为(5.85±1.97)、(6.19±1.88)%,均低于空白对照组的(9.89±1.86)%,差异有统计学意义(P<0.05)。苦柏坐浴颗粒组与马应龙麝香痔疮膏组肿胀度比较,差异无统计学意义(P>0.05)。给药6、12、24、48、72 h,马应龙麝香痔疮膏组和苦柏坐浴颗粒组大鼠肛周肿胀面积均小于模型对照组,差异有统计学意义(P<0.05)。致溃疡后第6、8、10天,苦柏坐浴颗粒组溃疡程度评分均大于模型对照组,差异均有统计学意义(P<0.05)。致溃疡后第8、10天,马应龙麝香痔疮膏组溃疡程度评分均大于模型对照组,差异有统计学意义(P<0.05)。结论苦柏坐浴颗粒具有抗炎、消肿、促进溃疡面愈合的作用。Objective To evaluate the efficacy of Kubai Zuoyu granule on the basis of anti-inflammation,detumescence and ulcer healing promoting,and to provide experimental basis for clinical evaluation for Kubai Zuoyu granule.Methods A total of 36 Kunming mice were randomly divided into blank control group,Mayinglong musk hemorrhoids ointment group,Kubai Zuoyu granules group,with 12 mice in each group.Vaseline,Mayinglong musk hemorrhoids ointment and Kubai Zuoyu granules were applied on the auricle of mice to make ear swelling test.After successful modeling,36 SD rats were randomly divided into model control group,Mayinglong musk hemorrhoid ointment group,Kubai Zuoyu granules group,with 12 rats in each group.0.5%carboxymethyl cellulose sodium(CMC-Na)solution,Mayinglong musk hemorrhoid ointment,and Kubai Zuoyu granules were applied to the anal swelling area at 6,12,24,48,and 72 h respectively to do the perianal swelling experiment.After ulcers,36 SD rats were randomly divided into model control group,Mayinglong musk hemorrhoid ointment group,Kubai Zuoyu granules group,with 12 rats in each group.Vaseline base,Mayinglong musk hemorrhoids ointment and Kubai Zuoyu granules were given respectively for perianal ulcer healing experiment.The anti-inflammatory effect of Kubai Zuoyu granule on mouse ear swelling,the anti-swelling effect on rat perianal swelling,and its effect on the healing of rat perianal ulcer were observed.Results The swelling degree of Mayinglong musk hemorrhoid ointment group and Kubai Zuoyu granules group were(5.85±1.97)and(6.19±1.88)%,which were lower than(9.89±1.86)%of blank control group,and the difference was statistically significant(P<0.05).There was no statistically significant difference in swelling degree between Kubai Zuoyu granules group and Mayinglong musk hemorrhoid ointment group(P>0.05).After 6,12,24,48,72 h of medication,the perianal swelling area of Mayinglong musk hemorrhoid ointment group were smaller than those of model control group,and the difference was statistically significant(P<0.05)
