参芪血脉通丸ICR小鼠经口给药急性毒性试验研究

摘　　要：目的:观察单次灌胃参芪血脉通丸ICR小鼠的急生毒性反应。方法:将ICR小鼠按体质量和性别随机均分为6组,分别为参芪血脉通丸8.0g/kg剂量组、6.0g/kg剂量组、4.5g/kg剂量组、3.4g/kg剂量组、2.6g/kg剂量组、2.0g/kg剂量组,每组10只,雌雄各5只。各组均以20mL/kg灌胃给药,给药前禁食14~16h,给药后观察6h,以后每天观察1次,持续观察14天。结果:参芪血脉通丸8.0g/kg剂量组、6.0g/kg剂量组、4.5g/kg剂量组、3.4g/kg剂量组24h单次给药,动物一般状态观察出现活动减少、呼吸加快或急促、步履不稳、低伏、俯卧不动、翻正反射消失、呼吸减慢或加深、抽搐、尿失禁、排稀便等症状,出现毒性反应的程度、数量与剂量相关,并出现与剂量相关的死亡情况。参芪血脉通丸2.6g/kg剂量组、2.0g/kg剂量组动物未见异常。在实验条件下,参芪血脉通丸的LD_(50)为6.2g/kg。结论:单次灌胃参芪血脉通丸可以引起ICR小鼠急性毒性反应。Objective:To observe the acute toxicity of ICR mice with a single intragastric administration of Shenqi Xuemaitong Pills. Methods: ICR mice were randomly divided into 6 groups according to body weight and gender, namely Shenqi Xuemaitong Pill 8.0 g/kg dose group, 6.0 g/kg dose group, 4.5 g/kg dose group, 3.4 g/kg Dose group, 2.6 g/kg dose group, 2.0 g/kg dose group, with 10 animals in each group, 5 male and 5 female. Each group were given 20 mL/kg intragastric administration, fasting for 14-16 h before administration, with observation for 6 h after administration, and observation once a day thereafter for 14 days. Results: Shenqi Xuemaitong pill 8.0 g/kg dose group, 6.0 g/kg dose group, 4.5 g/kg dose group, 3.4 g/kg dose group were given a single dose of 24 hours, and the general state of the animal showed decreased activity, accelerated breathing, unstable walking, low loft, prone motion, disappearance of righting reflex, slowing or deepening of breathing, convulsions, urinary incontinence, loose stools and other symptoms. The degree and quantity of toxic reactions were related to the dose, and appear to be related to dose-related deaths. There were no abnormalities in the 2.6 g/kg dose group and 2.0 g/kg dose group of Shenqi Xuemaitong Pills. Under the experimental conditions, the LD50 of Shenqi Xuemaitong Pills was 6.2 g/kg. Conclusion: A single intragastric administration of Shenqixuemaitong Pills can cause acute toxicity in ICR mice.
