摘 要:目的通过网络药理学方法筛选治疗宫颈癌(CC)的六君子汤主要活性成分及关键靶点基因,并分析六君子汤主要活性成分关键靶点基因的生物学功能。方法①运用TCMSP数据库、Swiss Target Prediction数据库筛选六君子汤的活性成分。②运用Drug Bank数据库、Uniport数据库筛选六君子汤主要活性成分的作用靶点基因,运用Gene Cards、OMIM、Uniport等数据库查询并获取CC的疾病相关作用靶点,利用Venny在线平台、String平台、Cyto-scape 3.7.2软件及其相关插件分析治疗CC的六君子汤主要活性成分作用靶点基因和CC疾病相关作用靶点基因的网络交互,筛选六君子汤治疗CC的关键靶点基因。③运用DAVID数据库(https://david.ncifcrf.gov/)进行基因本体功能及京都基因与基因组百科全书信号通路富集分析,分析六君子汤治疗CC的作用机制。结果六君子汤治疗CC的主要活性成分有槲皮素、山奈酚、柚皮素等149个。六君子汤治疗CC的关键靶点基因有促分裂原活化蛋白激酶3(MAPK3)、促分裂原活化蛋白激酶1(MAPK1)、信号转导及转录激活蛋白(STAT3)等30个。六君子汤治疗CC关键靶点基因主要参与的生物学功能为RNA转录、DNA链的特异性结合、细胞周期及细胞凋亡等,六君子汤治疗CC关键靶点基因主要富集在肿瘤相关信号通路、癌症中的蛋白多糖、ErbB信号通路等108条信号通路。结论六君子汤治疗CC的活性成分为槲皮素、山奈酚、柚皮素等149个,关键靶点基因为STAT3、MAPK1及MAPK3等30个。六君子汤活性成分关键靶点基因主要作用于RNA转录、DNA链的特异性结合、细胞周期及细胞凋亡等生物学过程,主要信号通路有肿瘤相关信号通路、癌症中的蛋白多糖、ErbB信号通路等。Objective To screen the main active ingredients and core target genes of Liujunzi Decoction in the treatment of cervical cancer(CC)based on the network pharmacology,and to analyze the biological functions of the main active ingredients and core target genes of Liujunzi Decoction.MethodsThe active ingredients of Liujunzi Decoction were retrieved by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)database and Swiss Target Prediction database.The target genes of the main active ingredients in the Liujunzi Decoction were screened by Drug Bank database and Uniport database,and the disease-related targets of CC were collected by Gene Cards,Online Mendelian Inheritance in Man(OMIM),Uniport and other databases.The network interaction between the target genes of the main active ingredients in Liujunzi Decoction and the disease-related targets of CC were analyzed by Venny online platform,String platform,Cytoscape 3.7.2 software and their related plug-in units.The core target genes of Liujunzi Decoction were screened.Signal pathway enrichment analysis of Gene ontology(GO)biological progcess and Kyoto Encyclopedia of Genes and Genome Encyclopedia(KEGG)were performed by the Annotation,Visualization and Integrated Discovery(DAVID)database.The mechanism of Liujunzi Decoction in treating CC was analyzed.ResultsThe main 149 active ingredients of Liujunzi Decoction in the treatment of CC were quercetin,kaempferol,naringin,etc.The obtained 30core targets for the treatment of CC by Liujunzi Decoction included mitogen-activated protein kinase 3(MAPK3),mitogenactivated protein kinase 1(MAPK1),signal transducer and activator of transcription protein(STAT3),etc.Those core target genes mainly participated in biological functions such as RNA transcription,specific binding of DNA strands,cell cycle and apoptosis,etc.The core target genes were mainly enriched in 108 signaling pathways,such as tumor-related signaling pathways,proteoglycans in cancer,and Erb B signaling pathways,etc.ConclusionsThe 149 ac
