摘 要:目的了解2015-2020年珠海市登革热的流行情况与病毒基因特征。方法收集登革热病例基本信息,采用RT-PCR对血清标本进行血清分型,阳性标本用RT-PCR扩增E基因并进行测序分型,获得的基因序列用MEGA6.0软件进行系统进化分析。结果2015-2020年珠海市共确诊登革热患者156例,其中131例患者的标本成功测序,包括DENV-1型96份,DENV-2型25份,DENV-3型10份;DENV-1型全部属于基因I亚型,分布在三个不同的分支上,这三个分支分别与柬埔寨、越南,斯里兰卡和柬埔寨、泰国等流行株进化距离较近;DENV-2和DENV-3病例主要为输入病例,与其对应的输入国流行株进化距离较近。结论近年来珠海市登革病热疫情为输入性病例引起的本地暴发流行,需加强与东南亚国家接壤的边境城市的登革热输入防控工作。Objective To analyze the prevalence and genetic characteristics of the dengue fever virus(DENV)in Zhuhai from 2015 to 2020.Methods Epidemiological data on cases were collected and analyzed,and serotypes of serum samples were identified using RT-PCR.Samples positive for the E gene were amplified using RT-PCR and typed via sequencing.The obtained gene sequence was analyzed using the software MEGA6.0.Phylogenetic trees of the E gene of DENV and reference sequences were constructed,their similarity was compared,and a phylogenetic analysis was performed.Results One hundred and fifty-six samples were positive for the DENV,and 131 of those were successfully sequenced.DENV-1 was found in 96 samples,DENV-2 was found in 25,and DENV-3 was found in 10.All of the DENV-1 strains belonged to subtype I and were distributed in 3 different clades designated A,B,and C.Clade A included 64 sequences that were genetically closest to reference sequences from Cambodia and Vietnam.Clade B included 2 sequences that were genetically closest to reference sequences from Sri Lanka.Clade C included 30 sequences that were genetically closest to reference sequences from Cambodia and Thailand.The sequence similarity between 96 DENV-1 and reference sequences was 95.16-99.53%.Cases of DENV-2 and DENV-3 were mainly imported,and DENV-2 and DENV-3 were genetically closest to the prevalent strains in the corresponding country.The strains of DENV prevalent in Zhuhai from 2015 to 2020 were not mutated.Conclusion In Zhuhai,local outbreaks of dengue fever are caused by imported cases.Efforts to prevent imported cases need to be enhanced in cities bordering tropical and subtropical countries.
