摘 要:为了探究氧化石墨烯(GO)对豆科植物抗病生理生态特征的影响及其作用机制,以紫花苜蓿(Medicago sativa)为材料,采用盆栽实验,将不同质量的GO添加到土壤中,使其质量分数分别为0.2%、0.4%和0.6%,测定种植60 d后紫花苜蓿的生长指标、根瘤数量、植物抗病相关酶活性和抗病物质含量.结果发现:GO处理显著降低了紫花苜蓿的地上干质量;0.4%和0.6%GO处理组中的植物株高显著低于对照组的株高.与对照组相比,0.4%和0.6%GO处理组中的苯丙氨酸解氨酶(PAL)活性显著降低,0.6%GO处理组中几丁质酶的活性显著低于对照组的活性.随着GO质量分数的增加,富含羟脯氨酸糖蛋白(HPRG)含量显著降低,0.6%处理组中的含量最低.0.4%和0.6%GO处理组中的木质素含量显著低于对照组的含量.由此可见,土壤中较高质量分数的GO(≥0.4%)能够显著抑制紫花苜蓿的生长,降低其抗病相关酶的活性和抗病物质的含量,增加紫花苜蓿的染病风险.In order to explore the effects of graphene oxide(GO)on the disease resistance characteristics of Leguminosae plant and its mechanism,a pot experiment was conducted by using Medicago sativa as the subject.Different mass of GO was added into soil,and the final mass fractions of GO were 0.2%,0.4%and 0.6%respectively.Some parameters including plant height,dry weight,root length,nodule number,as well as activity of disease resistant enzymes and contents of disease resistant substances were measured.The results showed that GO treatments significantly decreased dry weight of M.sativa.A significant decrease in plant height was observed under 0.4%and 0.6%GO treatments.Compared with the control,0.4%and 0.6%GO treatments significantly decreased phenylalanine ammonia lyase(PAL)activity.Under 0.6%GO treatment,chitinase activity was significantly lower than that in the control.With the increase of GO mass fraction,the content of hydroxyproline-rich glycoprotein(HPRG)significantly decreased,which was minimum in 0.6%GO treatment.Lignin contents of alfalfa were lower in 0.4%and 0.6%GO treatments than in the control.Overall,high mass fractions of GO in soil(≥0.4%)could significantly inhibit the growth of M.sativa,reduce the activity of disease-resistant enzymes and the content of disease-related substances,and increase the potential risk of infection.
