摘 要:目的:探讨柚子皮挥发油(PPVO)对于局灶性脑缺血再灌注损伤的影响。方法:40只健康雄性大鼠按随机分为假手术(SH)组、模型(NS)组、甘油溶剂组(GL)组、低剂量(LP)组(10 mg/kg)和高剂量(HP)组(30mg/kg),n=8/组。SH组仅行手术操作,余各组均建立短暂性大脑中动脉堵塞/再灌注(tMCAO/R)模型,并于再灌注时立即腹腔注射药物。观察再灌注24 h后神经功能学评分,通过2,3,5-氯化三苯基四氮唑(TTC)染色检测梗死面积,HE染色、尼氏染色观察病理形态学变化,免疫荧光染色观察星形胶质细胞标志物酸性胶质纤维蛋白(GFAP)和小胶质细胞标志物离子钙接头蛋白1(Iba 1)的表达情况。结果:再灌注24 h后,与NS组相比,HP组大鼠NDS明显下降,梗死面积显著减小,病理损伤改善,正常神经元明显增多,胶质细胞激活减少(P<0.05)。结论:PPVO(30mg/kg)可减轻局灶性脑缺血后病理损伤,减少梗死面积,并抑制星形和小胶质细胞的激活。Objective:To investigate the effect of pomelo peel volatile oil(PPVO)on astrocytes and microglia cells in rats after focal cerebral ischemia reperfusion injury(CIRI).Methods:Forty healthy male SD rats were randomlydivided into five groups(n=8/group):sham-operated(SH)group,model(NS)group,glycerol(GL)group,low-dose PPVO(LP,10 mg/kg)group,and high-dose PPVO(HP,30 mg/kg)group.The Sham group only performed surgery,and the other groups rats were established the model of transient middle cerebral artery occlusion/reperfusion(tMCAO/R).PPVO was administrated intraperitoneally at the time point of restore reperfusion,immediately.After 24 h of reperfusion,neurological deficit score(NDS),Hematoxylin-eosin(HE)staining,and Nissl staining were observed.Infarct volumes were detected by2,3,5-triphenyltetrazolium chloride(TTC)staining.The expression of astrocyte marker GFAP and microglia marker Iba 1 were detected by immunofluorescence(IF)staining in each group.Results:Compared with those in NS group,PPVO(30 mg/kg)had significantly decreased NDS and infarct volumes,improved pathological damage,increased nissl bodies,and inhabited the activation of astrocytes and microglia(P<0.05).Conclusion:PPVO(30 mg/kg)canimprove the pathological damage,decrease infarct volumes,and inhabit the activation of astrocytes and microglia.
