摘 要:目的观察平喘颗粒对哮喘大鼠气道重塑及TGF-β1/Smads信号通路的影响。方法40只Wistar大鼠,随机分为对照组、哮喘组(OVA)、平喘颗粒组(OVA+平喘颗粒)、地塞米松组(OVA+地塞米松),每组10只。应用OVA致敏液[10 mg OVA+100 mg Al(OH)_(3)]制备慢性哮喘大鼠模型。采用Masson染色观察肺组织病理变化,计算机图像分析系统(Image-Pro Plus Version 6.0)检测气道病理变化,蛋白质印迹法(Western blotting)检测肺组织TGF-β1、Smad2和Smad3蛋白表达。结果与对照组比较,哮喘组大鼠WAm/Pbm、WAi/Pbm明显增高(P<0.05),肺组织中TGF-β1、Smad2和Smad3的表达水平明显增高(P<0.05);与哮喘组比较,平喘颗粒组和地塞米松组大鼠WAm/Pbm、WAi/Pbm明显下降(P<0.05),肺组织中TGF-β1、Smad2和Smad3的表达水平明显下降(P<0.05)。结论平喘颗粒可以改善哮喘气道重塑,其机制可能与调控TGF-β1/Smads信号通路有关。Objective:To observe the effect of Pingchuan Granules on airway remodeling and TGF-β1/Smads signal pathway in asthmatic rats.Methods:40 Wistar rats were randomly divided into 4 groups:control group,asthma group(OVA),Pingchuan Granules group(OVA+Pingchuan granule),and dexamethasone group(OVA+dexamethasone),10 rats in each group.OVA sensitizing solution[10 mg OVA+100 mg Al(OH)_(3)]was used to prepare rat model of chronic asthma.Masson staining was used to observe pathological changes in lung tissue and computer image analysis system(Image-Pro Plus Version 6.0)was used to detect pathological changes in airways.Western blotting was used to detect the protein expression of TGF-β1,Smad2 and Smad3 in lung tissue.Results:Compared with the control group,WAm/Pbm and WAi/Pbm in the asthma group were significantly increased(P<0.05)and the expression levels of TGF-β1,Smad2 and Smad3 in the lung tissue were significantly increased(P<0.05);compared with the asthma group,WAm/Pbm and WAi/Pbm of the rats in Pingchuan Granules group and dexamethasone group were significantly decreased(P<0.05),and the expression levels of TGF-β1,Smad2 and Smad3 in the lung tissue were significantly decreased(P<0.05).Conclusion:Pingchuan Granules can improve asthmatic airway remodeling,and its mechanism may be related to the regulation of TGF-β1/Smads signal pathway.
