摘  要:目的基于网络药理学探讨温胆汤治疗哮喘作用机制。方法基于TCMSP数据库获取温胆汤中主要药物的化学成分及其对应靶点,基于OMIM、Gene Cards数据库获取哮喘相关靶点,利用Cytoscape3.6.1软件构建中药-靶点网络及应用STRING平台构建蛋白相互作用网络(PPI),应用R语言软件对关键靶点进行GO富集分析和KEGG通路富集分析。结果共获得52个化合物,131个药物与哮喘相关靶点,主要化合物为luteolin、beta-sitosterol、naringenin、Stigmasterol等。PPI网络中核心基因为Akt1、MAPK3、IL-6、VEGFA、FOS、JUN、MAPK1等;GO功能富集后富集数目较多的有ubiquitin-like protein ligase binding、DNA-binding transcription activator activity、RNA polymeraseⅡ-specific、phosphatase binding等;KEGG富集通路分析显示温胆汤治疗哮喘主要通路有PI3K-Akt signaling pathway、Endocrine resistance、Apoptosis、cAMP signaling等;KEGG通路与基因之间的网络图中关联数量较多的基因主要有MAPK3、MAPK1、Akt1等。结论本研究通过网络药理学对温胆汤治疗哮喘的作用机制进行探讨,为后期实验研究提供探索依据。Objective:To explore the mechanism of Wendan Decoction in the treatment of asthma based on the network pharmacology.Methods:The chemical components and corresponding targets of Wendan Decoction were obtained based on TCMSP database;the asthma related targets were obtained based on OMIM and Gene Cards database;Cytoscape 3.6.1 was used to build the traditional Chinese medicine target network;STRING platform was used to build protein interaction network(PPI);R language software was used to carry out go enrichment analysis and KEGG pathway enrichment analysis on key targets.Results:A total of 52 compounds and 131 drug-related targets were obtained.The main compounds were luteolin,beta sitosterol,naringenin,stigmasterol,etc.The core genes in PPI network were Akt1,MAPK3,IL-6,VEGFA,FOS,JUN,MAPK1,etc.;after enrichment of GO function,ubiquitin-like protein ligase binding,DNA binding transcription activator activity,RNA polymeraseⅡ-specific,phosphorase binding,etc.were more abundant;KEGG enrichment pathway analysis showed that the main pathway of Wendan Decoction in the treatment of asthma was PI3 K-Akt signaling pathway,Endocrine resistance,Apoptosis,cAMP signaling pathway,etc.;the major genes in the network map between KEGG pathway and genes were MAPK3,MAPK1,Akt1,etc.Conclusion:This study has explored the mechanism of Wendan Decoction in the treatment of asthma through the network pharmacology,and provided the basis for later experimental research.
