摘  要:目的分析利拉鲁肽对糖尿病大鼠体重、血糖以及鼠脑的影响。方法用链脲佐菌素(STZ)诱导SD大鼠产生胰岛素抵抗构建2型糖尿病模型,腹腔连续注射利拉鲁肽[0.3 mg/(kg·d)]6周,每周测量大鼠体重与血糖,注射6周后,测定鼠脑ANG-2,HMGB-1表达含量以及微血管密度(CD34)。结果在利拉鲁肽治疗组中,在第1周到第3周末大鼠的体重降低,3周后对体重的控制出现差异;利拉鲁肽对血糖影响存在个体差异,表达不一。与正常对照组相比,糖尿病组大鼠鼠脑内的ANG-2和HMGB-1表达增加(P<0.05);与糖尿病组大鼠相比,利拉鲁肽治疗组的大鼠鼠脑中ANG-2和HMGB-1表达降低(P<0.05)。与正常对照组相比,糖尿病组大鼠大脑中CD34表达下降(P<0.05);而利拉鲁肽治疗组的大鼠鼠脑与糖尿病组大鼠鼠脑相比,其CD34表达升高(P<0.05)。结论利拉鲁肽能在短期内降低糖尿病大鼠的体重;利拉鲁肽通过影响ANG-2、HMGB-1和CD34的表达,可能对糖尿病大鼠大脑起到预防和保护的作用。Objective To analyze the effect of liraglutide on the brain, weight and blood glucose of diabetic rats. Methods STZ-induced insulin resistance in SD rats resulted in type 2 diabetes mellitus. After continuous intraperitoneal injection of liraglutide [0.3 mg/(kg·day)] for 6 weeks, immunohistochemical staining and Western blot staining were used to analyze the expression levels of ANG-2, HMGB-1 and CD34 in rat brains, the weight and blood glucose of the diabetes rats were measured. Results In the treatment group of liraglutide, the weight of rats decreased from the first week to the third weekend, and the weight control was different after three weeks, while the effect of liraglutide on blood glucose was affected by individual differences, with different expressions. Compared with the control group, the ANG-2 and HMGB-1 expression levels in the brains of diabetic rats were increased(P<0.05), while the ANG-2 and HMGB-1 expression levels were decreased in the liraglutide group(P<0.05).Compared with the normal control group, the expression of CD34 in the brain of rats in the diabetes group was decreased(P<0.05), while the expression of CD34 in the brain of rats treated with liraglutide was increased(P<0.05). Conclusion Liraglutide could reduce the weight of diabetic rats in a short time. Liraglutide may play a preventive and protective role in the brain of diabetic rats by affecting the expression of ANG-2, HMGB-1 and CD34.
