摘 要:目的探讨儿童手足口病(HFMD)患儿急性感染期外周血中髓系抑制性细胞(MDSCs)的变化及临床意义。方法收集24例HFMD患儿和8例对照组外周血(PBMCs)标本进行研究。用流式细胞术测定MDSCs细胞及其亚群的比例,采用T细胞增殖实验评价HFMD患儿来源MDSCs的免疫抑制功能,评估HFMD患儿MDSCs比例与疾病炎症指标的相关性。结果与健康对照组相比,HFMD患儿PBMCs中多形核(PMN)-MDSCs显著升高(P<0.05),同时HFMD患儿来源PMN-MDSCs具有显著的T细胞增殖抑制功能。并且PMN-MDSCs水平与机体白细胞含量和中性粒细胞含量呈显著正相关关系,提示其与疾病进展密切相关。结论HFMD患儿急性感染期外周血中PMN-MDSCs升高,并具有显著的免疫抑制作用,是HFMD患儿潜在的细胞免疫治疗靶点。Objective To investigate the changes and clinical significance of myeloid inhibitory cells(MDSCss)in the peripheral blood of children with hand,foot,and mouth disease(HFMD).Methods Collect peripheral blood samples(PBMCs)from 24 children with HFMD and 8 healthy controls.Flow cytometry was used to determine the proportion of MDSCss.T cell proliferation experiments were used to evaluate the immunosuppressive function of MDSCss derived from children with HFMD.The correlation between the proportion of MDSCss with HFMD and disease indexes was evaluated.Results Compared with the healthy control group,polymorphonuclear(PMN)-MDSCs in PBMCs of children with HFMD were significantly increased,the difference was statistically significant(P<0.05),and PMN-MDSCs from children with HFMD had a significant T cell proliferation inhibitory function.Furthermore,PMN-MDSCs levels were significantly positively correlated with the white blood cell and neutrophil content,suggesting that it is closely related to disease progression.Conclusion PMN MDSCss in peripheral blood of HFMD children with acute infection increased and had significant immunosuppression,which is the potential target of cellular immunotherapy in HFMD children.
